A new domestic animal, alpaca,has been introduced from outdoor. Its unique economic, social and environment effects lead to the extensive attention. But alpaca's slowly and unpleasant reproduction is still a restrict factor for large-scale expand. How to solve this problem is a focus of academic circles,and the research of reproductive hormone is the most importance of the focus.Prolactin is one kind of polypeptide hormone and is mostly excreted by prehypophysis.It has been testified that have 300 or more functions.The purpose of this research is to research the partial prolactin(PRL) sequence of alpaca. According to conservative domain of the published sequence of prolactin gene of vertebrate, a pair of primers that can amplify alpaca PRL gene was designed and synthesized. The genome DNA was extracted by chloroform / iso- amyl alcohol and the tatol RNA by Trizol.A single DNA or cDNA fragment of about 1100bp or 510 bp ere amplyfied by PCR and RT-PCR individually, The products were purified used the reageat made by ourselves. The sequence was compared with published prolactin genes in Genebank by BLAST, the sequence showed hightest homology with PRL gene.And the highest homology is 97%.The result of research is of great significance to obtaining complete sequence of nucleotide ,extaction and purification of aPRL.
