Our country has the largest rice area in the world, the per unit area yield and the total output of rice is situated the world first place continuously. At the same time our country has in the world the most advanced hybrid rice breeding technology , large amount of hybrid rice was exported to foreign countries each year. It is estimated, in the next 10 years to 15 years, the world sown area of hybrid rice will amount to 100 million hectares, the hybrid rice seed demand is about 1.5 million tons to 2 million tons every year,Approximately some 12 billion to 16 billion US dollars markets spaces.Therefore, establishing a fast, accurate examination method to appraise the purity of hybrid rice varieties, and breeding hybrid rice varieties of high production, high quality, strong desease resistance are imminent, watershed.This article has summarized hybrid rice' s exportation condition, pretended the harm of the fake hybrid rice seeds, reviewed purity appraisal methods of hybrid rice and their good and bad points, the present using situation of Dongxiang wild rice in disease-resistant breeding. Introduced with emphasis the characteristic, discovery and examination methods of SNP(single nucleotide polymorphism) , the principle of real-time fluorescence PCR, TaqMan probe as well as rice molecular biological research progress of desease-resistance.In the experiment, we first designed primers to expand ITS (internal transcribed spacer) 2 subsequence of rice ribosome DNA.
