China has the largest rice area in the world. The per unit area yield and the total output of rice is situated the world first place continuously. At the same time our country has the most advanced hybrid rice breeding technology in the world. Large amount of hybrid rice was exported to foreign countries each year. It is estimated, in the next ten years to 15 years, the world sown area of hybrid rice will amount to 100 million hectares. the hybrid rice seed demand is about 1.5 million tons to 2 million tons every year, Approximately 12 billion to 16 billion US dollars markets spaces.In the research of hybrid rice's examination, we used to apply traditional methods, such as: the seed shape, the phenol dyeing, the seedling appraisal, field-plot planter method and so on. But these kinds of methods consume too much time, and the efficiency is very low. Moreover, it need strong experience, does not suit the quick test. In recent years, the PCR technology obtain the widespread application in the quarantine examination domain. people used the technology of DNA fingerprint analysis enhanced the examination speed and efficiency greatly, but this method needs to carry on a series of following processing, such as: the electrophoresis, the dyeing, the ultraviolet transmission observation and so on. These operations is tedious and must contact the carcinogen, which is easy to create the PCR product pollution in the laboratory. Another disadvantage is that we can only test molecular weight by judge the electrophoresis DNA fragment, and can not get more information. Therefore, establishing a fast, accurate examination method to appraise the purity of hybrid rice varieties is imminent.Real-time fluorescent PCR is a kind of fast, keen, safe examination method, which is used in the examination of bacterium, fungi, virus, gene modified organism recently.
