Method: IgG was purified from serum of giant panda and antiserum was produced. Single Radial Immuno-diffusion method to measure IgG was established on purified IgG and antiserum. Rerults: IgG was highly purified by saturation ammonium sulphate-DEAE ion exchange chromatography method, and the purified IgG was proved to be electrophoresis purification by SDS-PAGE; Antiserum was obtained after using purified igg to immunize rabbits three times; The antiserum was detected by the agar double diffusion method; the titer is 1: 32 and the specificity of the antiserum was good; The best dilution of the antiserum was 1: 30 and the best dilution of the sample was 1 : 10 ; The working rage was from 0.226mg/mL to 3.116mg/mL. Sensitivity was 0.113mg/mL; The recovery was between 103.1%~114.4%; The IgG concentration values of serum which belonged to 1-20 years old pandas was from 4.23mg/mL to 23.15mg/mL; The IgG concentration in milk which was secreted from 1 day to 19 day is from 3.54mg/mL to 14.18mg/mL.Conclusions: single immunodiffusion method can determined stably igg in serum and colostrums. IgG concentration in adult panda serum significantly higher than sub-adult panda. IgG concentration in panda milk decrease following postnatal days.
