| The genetic polymorphisms of 100 NanJiang meat goat and 30 LiaoNing cashmere goat was analyzed by microsatellite, PCR-SSCP, PCR-RFLP and sequence analysis.The effective markers which correlate with little size were fended ,as well as provide some DNA level basis for selecting and breeding prolific NanJiang meat goat. The study results were as follows: (1) 57 allelic were got in the 9 microsatellite and the average number was 6.3 and the most was 8, the least was 5. The PIC of 9 microsatellite loci all reached high polymorphisms (PIC>0.7), The heterozygosity was between 0.7344-0.8360.All these identified that Nan Jang meat goat have abundant polymorphisms and the potential of selecting and breeding were very large in Nan Jang meat goat. (2)13 markers from 7 microsatellites had been detected the significant positive correlation with the little size (P<0.05): OarAE101(109bp),BM1329(178bp), LSCV043(142bp,118bp),OarHH35(124bp,146 bp,104 bp,118 bp),TGLA68(117bp,133bp, 127bp),OarHH55(133bp,119bp), the significant negative correlation with the little size (P<0.05)was OarAE101(113bp),BM1329(210bp),LSCV043(136),OarHH35(108bp,114bp),TGLA68(103bp,123bp),OarHH55(151bp). (3)Three genotype (AA ,AB, BB)were discover in GDF-9exon1.The frequency of AA was 47% in Liao Ning cashmere goat. In Nan Jiang meat goat, the frequency of genotype BB was 38%, the frequency of genotype AB was 38% was 49%. The frequency of allelic B was 63% in NanJiang Meat Goat and The frequency of allelic A was62% . In the analyse of gene LH? , the frequency of allelic A and B were 46%and 54% in Nan Jiang meat goat , the frequency of allelic A was 40% in Liao Ning cashmere goat . (4) In gene LH?, the least squares means of genotype BB was significant higher than genotype AB and AA (P<0.05). In gene GDF-9, the least squares means of genotype BB was the most and the least squares means of genotype AA was the least .So the allelic B of gene LH? and GDF-9 was significant positive correlate with the little size. (5) The sequencing of LH? gene exon 2 that there was a mutation locus :T→C, GDF-9 gene exon 2 has a mutation locus: A→C too . But these mutations don't lead the change of amino acid sequence. |
