A Study On Embryo Abortion Of Chinese Jujube

Five excellent cultivars of Chinese jujube, including 'Lizao', 'Dongzao', 'Lajiaozao', 'Wudeng' and 'Xiangfenmuzao' were employed to establish the optimized culture system of early immature embryo, in addition to investigate the growing characters and abortion of embryo for 100 cultivars .The anatomy characteristics of seed development in 'Lizao' 'Buyuyihao' and 'Dongzao' were studied to reveal for clearing the characters and mechanism of embryo abortion. The effect of artificial pollination and spraying hormones on seed development also studied. The main results were summarized as follows:1 The ovule germination system of 'Lizao'. And the optimized culture system for embryo age on 'Dongzao' and 'Lajiaozao' were established.'Lizao' ovule germination medium was basic medium B5+ 1g/LAC and B5+IBA1.0 mg/L +BA0.4 mg/L,and the medium for better callus formation of ovule was B5+IBA 1.0 mg/L +BA0.4 mg/L.'Dongzao': starting system was MS+IAA2.0 mg/L +BA0.5+ LH 0.8g/l + sucrose 30g/l+agar4g/l+AC2g/l. Rooting medium was MS+IBA0.3~0.5mg/L+sucrose 30g/l+agar 4g/l. The optimal plantlet formation rate can reach 40 % under above condition.'Lajiaozao': starting system was MS+IAA2.0 mg/L +GA30.4 mg/L +LH 0. 5g/L+AC 1g/L+sucrose30g/L+agar4g/L. The bearing bud medium was MS+BA0. lmg/L +IBA0.5mg/L +NAA0.1mg/L +LH0.5g/L+sucrose50g/L+agar4g/L. Remult iplicated medium was MS+IBA1.5+BA1.0+sucrose30g/l+agar4g/l. The making ro ot medium was MS+IBA0.7 mg/L + sucrose20g/l+agar4g/l.2 Different plant growth regulator combination, embryo age and innoclation pose were the key factors in embryo culture . low level BA and high level IAA were beneficial to germination, but the probability of producing deformed seeding may be increased if adding too much IAA. when ovule was inoculated inverted or vertically, ovule germination rate reached 100%, Adding proper quantity LH was helpful to embryo germination, but may cause root deformation. The culturing temperature of 18-21℃ was fit for embryo germination.3 The seed rates were surveyed on 100 excellent cultivars of Chinese jujube at different growing periods of fruit.
