As one of the major crops in China and all over the world, wheat plays very important role in the development of Chinese national economy and social stability. Utilization of heterosis is one of important and useful ways to promote crop output and quality. Male sterility with Aegilops Ventricosa cytoplasm has been proven to be promising in hybrid wheat breeding, which has broad spectrum of restoring gene resources and has no distinct harmful cytoplasmic effect. While most of its restorer lines have no higher restoring ability, so it is impossible to use them directly in hybrid production. Breeding of good restorers is one of the prerequisite to produce hybrids using V-CMS system. Inheritance studies of restoring genes are theoretical basis of breeding restorers. DNA molecular markers assisted selection will greatly improve efficiency and accuracy of restorer breeding. At the same time, we may locate Rf genes on chromosomes and their arms by screening molecular markers linked to them, and this may provide a basis for cloning Rf genes. Here, we studied the inheritance of major restoring gene in two restorer lines of LK783 and JN13, mapped the major Rf genes in LK783 by SSR and ISSR markers. The results are as follows: (1) In mapping of fertility restoring gene in restorer line LK783 for V-CMS using molecular markers, we found that Xgwm11,Xgwm18,Xgwm273,Xgwm264 and UBC845-800 were linked to the restoring gene in LK783. They were located on chromosome 1BS by amplifying the DNA of nulli-tetrasomics and ditelosomics of CS with the 4 pairs of SSR primers and one ISSR primer.
