1575A,a new cytoplasmic male sterility material from of Canada in the experimenthas stabile sterility and maintainer lines from almost all the brassica napus l.,but its restorerline is more difficult to find.The feature is similar to ogu CMS(no minus green),while there isobvious difference between the two CMS lines,the seedings of1575A have grown well andhave normal nectarium instead of becoming yellow when the lobus cardiacus in lowtemperature.The rearerch group cloned the genes orf138and orf222in1575A by PCR andinfered the both male sterility gene of ogu and nap CMS in the1575A.Though a large numberof testcrossing,the restorer1601Rfo was screened preliminary,but the process of microsporedevelopment and the restoring gene were not studied deeper.In order to determine the sterile type of the1575A, paraffin section method inBrassicanapus L was used to observate and compare and pollen abortion periods andcytological characterics of five cytoplasmic male sterile lines and comparison of1575A withother male sterility in the similarities and differences,including pol,nap,shan2A types andtheir maintainers,the observation of anther development was made by means of paraffinsection method in Brassica napus L. in this study. At the same time,testing the restoring geneof1601Rfo by using the specific primers of ogu CMS restore gene. the results of the studyshow that:1.The results indicated that the pollen development process of1575A is differentfrom the other several sterile types.The anther abortion period of1575A CMS started fromtetrad to mononuclear stage. It was hard to release microspores from tetrad,and degenerationof tapetum was delayed. The anther development of shan2A,nap and pol CMS was inhibitedat the sporogenous cell,besides,a few pollen sacs could develop and produce normal pollengrains. The anther abortion of CMS758A started also at the sporogenous cell stage. However,it couldn′t produce sporogenous cell,the pollen sac and no pollen grains. Inconclusion,CMS758A aborted completely.2.The research used specific primers aleady announced and marked the restoringgene of ogu CMS by other people.There have two pairs of primers with special band bymarking the restoring gene of1575A,the primers is BnRFo-AS2F/BnRFo-AS2R and C02-f1/C02-rev1.The genetic distances between the two primers to target gene is11.1cM and6.5cMrespectively.Contrast,the primers P1and P2to the restoring genes of ogu CMS in molecularmarker,the restoring gene of1575A may originate from Rfo,...
