| Zoysia matrella, also called manilagrass, has many attractive characteristics, such as strong rhizomes and stolons, fine texture and broad adaptability, strong resistance, rapid growth, long green period, endure to trample and also elasticity. Due to its high turf value and horticultural value, it is widely utilized for green land and playground. To develop a tissue culture technique will be useful for propagating superior material and genetic improvement of the species.By taking stolon pieces as explants, we studied tissue culture techniques of Zoysia matrella. The main results are as follows:(1) The study disscussed factors such as explants sterilization, collecting time period and treatment. When sterilized with 70% alchol(45s)+0.1%HgCl2(8min), the frequency of contamination and browning was very low,only 7.52% and 9.73 % .Through comparison,the stolon pieces with a bud collecting in early spring to early summer were optional explants.(2) The axillary's buds bulged after 6-8 days and burgeoned after 15-20 days, then dark green bud clump formed. The best initiation and multiplication medium for the axillary's buds is MS+0.2mg/LIBA+2.0mg/LBA, the differentiation norm of sprouts is 6.57, and the average height of seedling is 4.8cm. Significant differences was observed.(3) The best medium for rooting is MS+0.1mg/LNAA+0.3mg/LIBA.When the seedling was incubated in the rooting medium for about 10-15 days, the white adventitious roots formed continuously. After 30 days, the average norm of rooting is 12.5, the average length of roots is 3.17cm, and the rooting rate is 100%.(4)The best culture medium for callus induction was MS+2mg/L2,4-D+0.2mg/LKT +200mg/LPro+500mg/LCH and the ratio of callus induction was 68.2%. The callus began browning after 16 days;(5)Light condition has predominant effect on callus induction.Under light,the callus is inducted 8.5 days and becomes brown after 12.5 days; Under dark, it takes 16 days induce callus and the callus become brown in 22 days. Significant differences was observed. |
