In this paper, we established a quantitative determination method for platelet activating factor (PAF), that is, washed platelet aggregation test (bio-assay), to explore the changes of the concentration of PAF before and after artificial fertilization in giant panda. At the same time, the activity of platelet activating factor acetylhydrolase (PAF acetylhydrolase, PAF-AH) was determined by the changes of substrate assay. So we analyzed the feasibility of early cyesiognosis by detecting the concentration of PAF and the activity of PAF-AH from giant panda plasma.Thin layer chromatography separation and platelet aggregation assay were suitable of the determination of platelet activating factor, the results were satisfactory in case of standardized operation. The recovery rate of PAF (throughout thin layer chromatography separation) was 96.3%. Range of linearity was 2~10ug/L, rabbit platelet optimum numbers:3.5~4.010 11/L.When estrogen peak in female giant pandas' urine sample comes a sudden drop, we started giant pandas' mating by nature mating or artificial fertilization (AI), and collected the plasma through to the last AI. PAF released by fertilized ova or early embryo can result in the decrease of number in the peripheral platelets, so giant pandas, which the number of platelet decrease before and after mating, were grouped to the group with decreasing, and the other to the group without decreasing. The result showed that the concentration of PAF in plasma on the second day after mating was not significantly between the two group[(3.85?.60) ug/L VS (3.79?.76) ug/L, P>0.05]; As far as in the group with40decreasing , the concentration of PAF before decreasing was not significantly than the concentration after decreasing[(3.78?.65) ug/L VS (3.85?.60)ug/L, P>0.05]. According having parturition or not, giant pandas were grouped to pregnancy group and no pregnancy group, the result showed the concentration of PAF on the second day after mating was not significantly between the two groups[(3.80?.78) ug/L VS (3.79?.76) ug/L, P>0.05].We detected the PAF-AH activity from the plasmas of two giant pandas with decreasing before and after mating, and found that all the PAF-AH activity after mating were lower than those before mating [(from12.42nmol.min-1.ml-1 to 10.27nmol.min-1.ml-1) and(froml3.33nmol.min-1.ml-1 to 12.11 nmol.min-1.ml-1)].Based on the above findings, we can conclude that detecting the concentration of PAF in giant panda plasma can't be used as a way of early cyesiognosis for giant panda, and we suggest trying the other way to diagnose early pregnancy. And the changes of the activity of PAF-AH indicated that the activity may be different during early pregnancy, and father research is necessary to understand the changes rule of PAF-AH activity in giant panda.
