Bordetella avium, an avian respiratory disease pathogen,causes an upper respiratory tract disease called bordetellosis ,which is a high contacted infectious disease in birds. And it is responsible for substantial economic losses to the poultry industry. It has been reported that the infection ratio of B.avium was 10~50% in parts of China, it caused much ecnomic loss in farms with delayed growth ,low feed reward of the stuffer (Zhang Shaoxue,1992;Wang Jingyu,1997;Zhuang Guohong,1998).In China, the pathogen can induce the death of embryo in hatching and the acute death of chicken after birth(Zhu Ruiliang,1991). Chicken,less than one-week,is most susceptive.The diseased chicken charactered by rhinotrachititis,depressing,food uptake reduce,and the most died of exhaust or infection by other pathogens.The chickens more than one-month have some resistance to this disease,and the mature chicks do not present symptom after infection,but the eggs can be contaminated,and it causes vertical spread of this disease.Because the traditional methods of microbe seperation and identification need much time and effort,it can not aquire exact result in 2~3 days.But the rapid identification technic can get over these deficiency. In this research,we explored the epidemiology of Bordetellasis in chicks and the heredity characters of different B.avium strains,and developed corresponding rapid detection methods with labelled antibody technique.It will be helpful in prevention of this disease.The research includes four parts:Part One :Survey of the Infection of Bodetella.avium in Parts of ChinaIn our study , serology test was operated with 801 samples collected from 25 poultry farms in GuangXi,HeNan ,ShanDong and AnHui province,and evaluation was done with Bodetella.avium(B.avium) infection ratio in pultry farms in China.The result of our survey indicates that the infection of B.avium has been existed widely in China. In the 801 samples,there are 326 which is from less than 70-day commercial poultrys,and 475 from more than 70-day parents poultrys .The result indicates that it is much more severe in more than 70-day age parents poultrys , with the everage infection ratio of 51.79%,and the infection ratio was 26.32%(65/247) in Guangxi, 75.63%(121/160) in He'nan ,87.3(48/55) in Shandong and 92.31%(12/13) in Anhui. But it is low in poultrys of less than 70-day age. Statistic analysis indicate,the infection ratio of B.avium is notabily diffent (p<0.05)in ages,but not diffent(p>0.05)in areas.Regress analysis indicate the infection ratio of B.avium is notabily positave correlation with ages(r=0.9282,p<0.01)。Therefore,it is necessary to think about the effection of age in evaluation of survey result. Part Two:The Chromosome G+C mol% Determination of B.avium Bordetella avium was a pathogen which can induce upper respiration tract disease in avian,it can cause much economic loss in pultry farms. The inability to distinguish isolates rapidlly has hampered generally investigating and prevented a complete understanding of the transmission mechanisms of this disease. In our research, The G+C mol% determination of five B.avium strains was operated,and the range was major in 61.9~62.9. The G+C mol% determination can be one of the basis and relative determinant in microbe identification. Therefore,the range of chromosome G+C mol% has important significance in identification of new separated strains.But, chromosome G+C mol% determination was mostly used in denial,because the different strain can has the same G+C mol% content(Han Wenyu,1992).Therefore,the identification of a new saprated strain,should make systemic examination.Part Three:Experimental Study in Detection of Bordetella avium by Direct Fluorescent-labelled Antibody TechnicAt present,the main methods of B.avium identification were microbe culture identification,antiserum agglutination,ELISA,seqence of 16S rRNA,PCR and immunofluorescence test,etc(Blackall P J,1984;Blore P J,1991; Tsai H J,1991;SureshP,1993;M.M.Kattar,2000).The method of imm...
