| The experiment was conducted by using of one-day-old chicks. the peripheral blood,thymus,bursa of Fabricus and spleen were sterilely removed for the culture of their lymphocytes. The T lymphocyte proliferative responses were determined, and supernatants from concanavalin A(ConA)stimulated cultures were assayed for the inductive activity of interleukine-2(IL-2). The B cells were stimulated with phorble myristate acetate(PMA)to determine the B lymphocyte proliferative responses. T lymphocytes IL-2 inductive activity and lymphocyte proliferation of chicks treated with HVEF were examined by using MTT method. The results were as follows:The T lymphocyte IL-2 inductive activity of thymus and spleen of positive HVEF chicks was significantly higher from 1 to 35 days than controls and negative HVEF chicks after treated with HVEF(P<0.05,P<0.01). By contrast, T lymphocytes IL-2 inductive activity of thymus and spleen of negative HVEF chicks was significantly lower compared with controls from 4 to 14 after treated with HVEF. The results showed that the secretion of IL-2 of thymus and spleen was augmented after treated with positive HVEF, so as to make molecular regulation of central and peripheral immune organs enhanced, whereas that of chicks decreased after treated with negative HVEF. The peripheral blood, thymus and spleen T lymphocyte proliferation were significantly enhanced from 1 to 21, 1 to 35 and 4 to 35 days , respectively, compared with controls and negative HVEF chicks after treated with positive HVEF(P<0.05,P<0.01). The peripheral blood, thymus and spleen T lymphocyte proliferation were significantly decreased from 4 to 21, 1 to 21 and 4 to 21 days , respectively, compared with controls after treated with negative HVEF(P<0.05,P<0.01).These results showed that the peripheral blood, thymus and spleen T lymphocyte function of chicks treated with positive HVEF was significantly elevated. By contrast, the peripheral blood, thymus and spleen T lymphocyte function of chicks treated with negative HVEF was inhibited.The peripheral blood, bursa of Fabricus and spleen B lymphocyte proliferative response to PMA were significantly enhanced from 1 to 35, 1 to 35 and 4 to 35 days , respectively, compared with controls and negative HVEF chicks after treated with positive HVEF(P<0.05,P<0.01). The peripheral blood, bursa of Fabricus and spleen B lymphocyte proliferative response to PMA were significantly lower from 4 to 21, 1 to 14 and 4 to 14 days , respectively, compared with control chicks after treated with negative HVEF(P<0.05,P<0.01). These results showed that the peripheral blood, central and peripheral immune organs B lymphocyte function of chicks treated with positive HVEF was significantly elevated. By contrast, the peripheral blood, central and peripheral immune organs B lymphocyte function of chicks treated with negative HVEF was significantly decreased.The results above disclosed comprehensively and firstly from cellular and molecular way that the effects of HVEF on immune regulation of cytokins and the peripheral blood and immune organs cellular and humoral immunity, in order to provide rational foundation for exploitation and utilization of HVEF. |
