Key factors in rose regeneration and transformation by pBinETRl are studied for new anti-senescence rose. The results show that:(1) Cultivar, auxin, explant, GA, ABA, condensate are the factors affecting rose callus induction. Cultivar and auxin are the main factors. 'Samantha' is best for callus induction, then one by one is 'Kardinal', 'Abans', 'Jimthyianky' and 'Babylon'. The results are not the same for inducing callus by using the different explants. The callus from petiole is good. Each cultivar has its proper explant and medium for callus induction. NAA is better than 2,4-D, the proper concentration is 2 mg/L-4mg/L. Adding ABA or GA don't affect the induction ratio, but the rigidity and color of callus can be improved, the proper concentration for ABA is 0.2mg/l and GA is 1.0-2.0mg/l respectively. Gelrite is better than agar for callus induction.(2) The Differentiation of rose callus is affected by cultivar, explant and cultural condition, in which the cultivar is the most important factor. One bud-like organ is differentiated from ' Samantha "s callus in MS+ BA2.0mg/L+ NAA1 .0mg/L. Leaves-like are differentiated from 'Samantha' callus in MS+ KT0.5mg/L+ the NAA2.0mg/L with liqulid medium of MS+TDZ0.05-0.1 mg/L. The bud and plantlet are differentiated from callus of 'Jimthyianky' in MS+ KT0.5mg/L+ the NAA2.0mg/1. 'Kardinal' has no differentiation yet. Appropriate dark condition is better for differentiation from yellow and fragile callus(3) Cultivar and antibiotic are the main factors for transformation, different liquid dilution rate, different co-culture time and different condensate do not affect the survival rate of callus. Different cultivar results in different callus survival rate by Agrobacterium tumefacience. 'Samantha' is better than 'Jimthyianky'. The proper concentration for Kan is 200mg/l-400mg/l and Car is 200 mg/L-400mg/L respectively.Above all, cultivar is the main factor for rose regeneration and transformation. In this research, callus are induced from 'Samantha', 'Kardinal', 'Abans', 'Goldlike' and 'Babylon', but only the callus of 'Samantha' and 'Jimthyianky' differentiate into bud-like organ, callus of 'Jimthyianky' also differentiate into bud and plantlet. The Kan-resistant callus of 'Samantha' and 'Jimthyianky' are acquired. This paper provided a good foundation for rose transformation in the further research.
