Through morphological markers, MDHâ…¡ isoenzyme markers and RAPDmolecular marker of four lines in A.m.ligusica, as genetic resources were comparedand analyzed.The results showed as follows:1 There existed significant differences (P<0.05) among adult workers of 4 lines in tongue length and right forewing area. Australia-Italian bee workers were the longest tongue and Ben-Yi bee workers the shortest. The right forewing area of Pinghu Jiang bee workers were the longest and Ben-Yi workers the smallest,And there existed no significant difference among bee workers of 4 lines in head weight. The results were different from the production capability corresponding to the four lines and demonstrated that individual morphological signs might not be regarded as a standard to evaluate population production. 2 The analysis, conducted by means of IEF PAGE, produce the flowing results iscc genotype had the highest genotypic frequency, Pinghu Jiang bee workers had the highest genotypic frequency ( ep=0.5425 ) , American-Italian bee workers had the lowest one. in gene frequency, c gene prevailed and Pinghu Jiang bee workers had the highest one, Australia-Italian bee workers the lowest.The purity degree of all lines surpassed 0.50, and Pinghu Jiang bee workers got a highest value of 0.82. The independence test indicated that differences between the 4 lines in genotypic frequency, gene frequency and hybrid-purity degree were statistically significant(P<0.01). The results demonstrated that c gene frequency might be regarded as an evaluating marker and c gene frequency might be adopted as a standard to evaluate the production of royal jelly.3 RAPD-PCR analysis of 30 random primers indicate that primer W18 (5'-CGGACGGCGG -3') and W23 ( 5'-GTACCGCCCG-3') leaded to polymorphie bands after amplification. 2353bp and 392bp all of W18 amplified bands were only in American-Italian bee workers, 2092bp band for high productivity of honey. 632bp was specific for high productivity of royal jelly. Of all W23 amplified bands, 651bp may be served as a specific band to distinguish Australia-Italian bee from other breeds. The-test results indicated that of gene frequencies polymorphic bands were significantly different among these four lines(P<0.01).4 The chaos -analysis by amplification using W18 and W23 primers showed that Pinghu Jiang bee and Ben-Yi bee might have a close kinship.
