To reveal the relationship between endogenous hormones and flower development and obtain correlative protein in specifically period of flower development as well as study the f mechanism of flowering in molecular level in Betula platyphylla Suk, the changes of endogenous hormones and protein expression during the flower development were analyzed, combining with the morphology and anatomy.The content of GA, ZR, ABA, and IAA during the development of male flower of this species was measured with ELISA in order to study the variation of the endogenous hormones in different stage of flower development. The result showed that low content of GA and ABA, IAA and ZR during the first stage of male flower development were useful to differentiation of male flower. High content of ABA, ZR and IAA was advantageous to differentiation of stamen primordium and the form of archesporial cell.The changes of protein expression during different period of female flower development were studied with SDS-PAGE. The best method for extracting solube proteins from female flower was selected and the program of silver staining for polyacrylamide gel was established. The result showed that there exists a differential protein band with 40.1kD during the first stage of inflorescence primordium and bract primordium's differentiation and development. Along with the inflorescence differentiation, the pistil primordial began to initiate and then went to dormancy, when there exists other differential protein bands with 23.5kD and 21.9kD.Two-dimensional gel electrophoresis was employed to analyze the protein patterns of male flower in the early period, middle period of inflorescence development as well as the formation of floral apparatus. An optimum method for 2-dimensional gel electrophoresis of male flower was set up, which the extraction buffer IV and improved TCA-acetone precipitation method were used to get protein, and immobiline DryStrip of pH 4-7 with protein from 50ug to 60ug was used to do electrophoresis, silver staining was used to get better 2-DE images. In the different periods, differential protein spots with 275, 197, and 135 in number were obtained respectively. Most of them are proteins with low molecular mass, which were probably related to the formation of flower meristem tissue and floral apparatus such as the formation of filament and anther respectively. |
