Retrotransposons exist widely in higher plants, being one of the main continents of plant genome. They are a kind of transponsons with the largest number, occupying 50%~80% maize genome, 90% wheat genome and 17% rice genome. Therefore, understanding the regulation rules of wheat retrotranspons is important for investigation of wheat genome. Retrotranspons with activation in wheat genome has not been reported so far.In this study, retrotranspons with activation in wheat was firstly reported. Based on the two conserved sequences of retrotranspons, two primers were designed. By the method of RT-PCR, a set of fragments containing the sequence of retrotranspons was amplified from the cDNA of wheat translocation line Pm97034 with resistance to powdery mildew. Sequence analysis of the randomly selected 23 clones suggested that they were highly heterogenous and divided into at least eight groups, which were tentatively named asTaRT-1 to TaRT-8. Southern blot hybridization indicates that they exist as multiple copies. Northern blot shows that TaRT-1 is only expressed at a low level under normal conditions in seedlings, but at a significant high level when induced by wheat powdery mildew, jasmonic acid and salicylic acid. And no expression of TaRT-1 was detected during the process of tissue culture. These results suggest that TaRT-1 is highly sensitive to abiotic and biotic stresses. The yield of wheat is greatly affected by the environmental stresses like low temperature, drought salinity salt and alkaline. More than half of the fields in China is droughty, half-droughty or salty and alkaline. And the losses caused by biotic stresses as diseases and insects significantly restrict the development in these areas. Therefore, investigation and breeding of stress-tolerent wheat varieties are important in practice.In addition, 6 distinct fragments were obtained in this study. Sequence analysis indicates that they are stress-tolerant related genes.
