Halothane gene is the main reason for porcine stress syndrome (PSS) and make obvious effects on meat quality o The homozyous abnormal pigs (HaTHal"), which is sensitive to the stress, can produce pale% soft and exudative meat (PSE) and can causes maior economiclosses in swine industry.In this thesis, the genotype frequencies of the halothane gene hi Chenghua pig> Yanan pig, Neijiang pig and Taihu pig of China native breed pigs were investigated by the PCR-RFLP technique, and the amplification products were analyzed in sequence polymorphisms .The results of restriction enzyme sites showed that the halothane genotype of all the 196 porcine cases, including 36 Chenghua pigs, 60 Neijiang pigs, 62 Taihu pigs, 30 Yanan pigs, wasNN.In the study,, 659 bp of the amplification products were sequenced in 16 individuals from five breeds including four china native breed pigs and one imported breed . Combining such 16 sequences with the five published sequences , the sequence analysis results indicated that there were 16 polymorphic sites with 3 transversion and 13 transitions in the 21 sequences and according to the base substitutions , 15 haplotypes were identified . In these polymorphic sites, the 18662 base of four Neijiang pig is all T, but other breeds have not the same mutations o T18662 was the breed character of Neijiang pigs and the similar mutation of the breed character were not found hi other breeds .All the sequences have the similar compstition of nucleotide . It was found that haplotype diversity and nucleotide diversity among these breeds and average number of nucleotide differences between populations ( k ) , average number of nucleotide substitutions per site between populations (Dxy) and number of net nucleotide substitutions per site between populations were very low. There were similar conclusion after computing mean distance within groups and between groups by kimura 2-parameter model .In 445-579 coding region, the amino acid variations were not foundo...
