With Phenylalanine Ammonia-Lyase (PAL) and Stilbene Synthase (STS) as the main indicators of grape resistance to disease, the effects of main components of E26 agent on the activities and genes' transcriptions of PAL and STS of suspension cells of two grape varieties (qiouhei and chixiazhu) were studied. The objective was to determine whether there existed induced disease resistance in the mechanisms of E26 agent controlling the grape crown gall.In the experiments, the medium for grape callus induction was screened and that for grape cell suspension cultivation was developed. Based on the comparison of several kinds of protocols, Fatima Ragueh's protocol was modified for grape UNA isolation.The results showed that, PAL activities were higher at most investigated hours after treated with the whole fermented liquid or metabolized liquid, while increased indistinctively and changed fluctuantly after treated with E26 cell or culture medium. Northern Dot Blotting expressed that the profiles of transcriptional time and quantity of pal had great similarities with those of PAL activities, and the transcription of sts is clearly coordinated with that of pal.The conclusion is that induced resistance may be one of the mechanisms of E26 agent controlling crown gall of grapevines in which metabolized products of E26 strain play the major role.
