Influence Of The Storage Conditions On The Seed Longevity Of Mustard (Brassica Juncea Coss) And Its Mechanism Research

In this experiment (Xinxuan Ercai, Yufeng Zhacai and Yuxuan Bangcai ) , sixteen different treatments of L16(45) positive-cross experiment designing with such three factor as temperature, moisture content of seed anchrelative humidity, were adopted to store the seeds of three mustard varieties.In this experiment, multinomial physiological and biochemical indexes relating to seed longevity, including percentage of germination % relative electrical conductivity and the content of sinapine in seed exudates, activities of SOD > POD, CAT, the content of MDA, protein, free fatty acid and dissolubility sugar of three kinds of mustard seeds in 16 storing conditions , were investigated respectively. At the same time , a series of physiological and biochemical changes of seeds in storing process were studied in a systemic way* through the aspects of seed viability, membrane permeability, membrane fat peroxidation and seed storing material. Furthermore, the mechanism influencing seed longevity under storage was discussed and some conclusions were drawn. First, relative electrical conductivity, activities of SOD* POD, CAT and protein content were reduced with the increase of the seed deterioration degree .Second, the change of dissolubility sugar content of seed was very complicated, as a whole, it seemed that there was not a distinct trend of increase or decrease, so it needs further research to testify the reliability of regarding dissolubility sugar content as an index to forecast seed longevity. Third, relative electrical conductivity, the content of MDA and sinapine in seed exudates added with the increase of the seed deterioration degree .Principal component analysis indicated that the correlativity among these indexes were distinct. There is a obviously positive correlation between the index of seed viability and the indexes of the activities of SOD, POD, CAT, MDA, protein content and dissolubility sugar, while there is a obviously negative correlation between the index of seed viability and the indexes of, relative electrical conductivity, the content of MDA > free fatty acid and sinapine, in seed exudates. Based on theseresults, we can conclude that all of these indexes, except dissolubility sugar, are not only the main factors influencing the seed viability but the-reliable physiological and biochemical indexes evaluating the seed longevity.Concerned with all varieties and indexes in a collective view, variance analysis indicated that the capability of influence of storing temperature, original moisture content of seed and relative humidity in decreasing order was temperature > relative humidity> moisture content of seed. Temperature had remarkable influence on each index of the three varieties. The result of principal component analysis showed that the treatments of No 15 (^BsC.*) and No 16 (A^B.^) were much better than other treatments stored seeds, while Nol5(temperarure 4癈, original moisture content about 8.5% ,relative humility 43%) was a little better than Nol6 (temperature 4癈, original moisture content about 6% ,relative humility 53%), but the result of variance analysis indicated that there was no prominent difference between the two treatments.In addition, the storing endurance of seeds with different genes was distinct, the sequence of endurance to sotre was Yufeng Zhacai>Xinxuan Ercai> Yuxuan Bangcai. In addition, different levels of each factor had different influence on seed longevity.The experiment results indicated that among the sixteen treatments of storing, All the seeds of each variety under the treatments of No1,No2,No3 and No4 lost the germination capability after three months storing, while other physiological and biochemical indexes were very different with the ones of seeds under other storing conditions, which showed that under condition of 37C, it was disadvantageous formustard seed to store under any level of moisture content and relative humidity in seed.Principal component analysis indicated that the mechanism that mustard seed became aged and deteriorated in...
