| The male sterile of plant is the basic of hybrid vigor. It is also the best material for the research of heredity of cytoplasmic , the interaction between nuclear and cytoplasmic and the development of pollen. So it has become the hot point of the research in plant heredity and molecular biology. It has showed that plant CMS is the result of the interaction between nuclear and cytoplasmic. genes in nuclear and cytoplasmic control the ability of fertilize together, but most of the researches are focus on heredity material in cytoplasmic. especially in mitochondrion. The research on how nuclear genes control the genes in mitochondrion is really less. For the reason of this problem, we use DDRT-PCR to study the difference of genes' expression. The material we use is T-type wheat cytoplasmic male sterile lines 75-3369A and their maintainers 75-3369B. We choose 14 different materials, which from 4 different times and 2 different organ. We use oligodT::CG as arbitrary primer and 8 random primers for PCR amplification. We select three better combinations. over3.640 cDVA fragments range from 100bp to 1000bp in length are obtained. Most of the fragments are the same between A and B, which showed that the heredity background of the materials we use is good. We also discover that the different fragments of three nuclei time are rather few. We use 63 fragments to Reverse-Northern blotting. And get 4 different fragments that have blot signal. We cloned them all. and have them to BLAST. Database searches indicate that 1 cDNA fragmentshow high homology to known genes, which mainly include MADS-box transcription factor. The other two cDNA fragments show high homology to Oryza saliva (3-5 leaf stage) EST. the last one is not similar to any gene or protein but it has a ORF , it may be a new protein. Implications of these results for the possible molecules in wheat salt tolerance are discussed. |
