Dendrobine was isolated and purified from the wild Dendrobium nobile Lindl. The relationship between growth and effective components accumulation was discussed, and It was also studied the the growth and effective compoments accumulation influenced by different factors.the main results were as following:The results of thin-layer chormatography and GC-MS showed that the crystal getting form the wild type Dendrobium nobile.was dendrobine. The purity was 96.558%.The growth curve of PLBs cultivate on solid culture medium was "S" type. There is an initial lag phase of twenty days at the beginning of the culture, followed by a rapid growth period of thirty days and The growth rate declined after fifty days. In the sixty days of culturing, the total amount of alkaloid and polysaccharides were increased with the increasing of biomass of PLBs. The contents of both alkaloid and polysaccharides reached peak after fifty days of cultivate, and then there was a decline in the last ten days of cultivate.PLBs on liquid stationary medium whose growth curve was "S" type too, and multiplied faster than those on solid medium.And the liquid medium It should that liquid stationary culture was more suitable for the accumulation of alkaloid and polysaccharides.The growth curve of PLBs cultivate(cultivated) in dark was also"S" type. However, compared with the ones in light, their multiplication rate was slowerbut there was a little increase on the content of alkoloide and great decline on polysaccharides. so , dark condition was unsuitable for the cultivating interested in the effective components of PLBs. But after about ten days of cultivation in dark ,the color of PLBs was light yellow and more disperse compare with those cltivated on lightBoth MJ and ABA can inhibit the growth of PLBs and increase the accumulation of alkaloid and polysaccharides. From this experiment,it can get the following the conclusion:PLBs cultivated on the medium concluding MJ25 u mol/L and 10 U mol^ ABA0.25mg/L and O.lmg/L can not only increase the accumulation of effective components notability.,but also restrain the growth of PLBs in lowest degree. |
