A regeneration system of tissue culture from Cotyledons of Spring onion and blood among some Allium lines were learned in this paper. The main results are as follow.(DThe optium medium and hormone comnination of callus induction from Cotyledons, subculture of callus, shoot regeneration from callus, shoot-extension induction and root induction are: MS+2,4-D 2.0 mg/L+KT 0.5 mg/L, MS + 2.4-D 2.5 mg/L+KTO.5 mg/L, MS+2,4-D 0+KT 1.0 mg/L, MS+KT 0.5 mg/L and basal MS medium.(2)The result of paraffin cut section to gloublas covering callus shows that a cluster of cells is the Primodium of shoot, with dense cytoplasm situate within it and the gloublas can regenerate new ones if they are subcultured.(3)Three plantlets of twenty two regenerated ones from tissue culture are variations which are different in stoma,the number of chromosome from root-tip somatic cells and external morphological characters. It shows that tissue culture can be thought as an effective method of making variations.(Where is a tetraploid plantlet among regenerated ones which is different in width, length and density of stoma and the number of chromosome compared with normal plantlets.(5)Twelve lines belonging to species of Allium were analyzed by RAPD markers to determine their blood relationship. Of the 20 primers screened, 10 showed consistent banding patterns and amplification and produced 65 bands with 51 polymorphic. Relationship between the lines determined by UPMGA cluster analysis demonstrated No.3 Welsh Onion and Jv Second Welsh Onion, Dianyi Welsh Onion and Laiwujitui Welsh Onion, No.5 Welsh Onion and Zhangqiu Welsh Onion were clustered together respectively which was the same as botany classification. So RAPD can be thought as an effective method to determine blood relationship.
