16 bud mutation primary election strains of 慒uji?apple and origin cultivars as materials the cytology such as pollen pattern character. stomata size and density were studied, and applied RAPD molecular marker technology identified these materials .In addition ,constructed three cultivars ISSR fingerprint. Results showed that: 1. Polkn size of every sport line was identical to the one of control cultivar, all of them were triangle ,the size and shape were regular, Stomata density and size of every bud mutation primary election line was same to those of contrast variety, 2. We established steady RAPD桺CR system .Amplification reaction volumes ~re 25 ~ I,~h amtaining 10mM TIis桯C1pHSJ3, 50mM KC1,2.OMmMgC12,0. 19mM each of dATP,dCTP,dGTP and dTTP,0.24 Li M prinier,3Ong DNA and IU Taq DNA polymerase (Promega). 3. The study chose 300 random primers, several random primers had distinguished 5 bud mutation lines from the control cultivar, respectively. 4. Constructed ISSR桺CR system .PCR amplification was performed in 25 u I containing 10mM Tris桯CI pH8.3,5OmM KC1,1.5mM MgC12,0.20 mM each of dATP,dCTP,dGTP and dTFP,0.40 L~ M priiner,2OngDNA and 1U Taq DNA polymerase(Promega). 5. Established the fingerprint of 慒uji?慡traking?慛ew Century?慉merica 8? 慛agafu2?慛agafu 6? 6. ISSR analysis was made in all bud mutation primary electionstrains .The primer 1S026 could identify nine bud mutation primary strains.
