Isolating,identifying,Sequencing And Analyzing Of Rubella Virus Epidemic Strain

Rubella virus is one of the virus that can lead to congenital birth defects. Rubella virus is a unique species belonging to genus Rubivirus of Togaviridae. Humans are the only known host of rubella virus. Rubella virus can infects human easily and it is spreaded by respiratory, resault in rubella in children and adult. Rubella virus infects unimmunized women during early stages of pregnancy ofen induces congenital defects in the newborn. In oder to investigate the relationship between the epidemic strain, the vaccine strain BRD II in our country and world strains. We detected 61 blood samples by reverse transcription and nested PCR. From the 7 samples, we gained I isolate, which was confirmed by cytopathic effect(CPE), hemagglutination, electron microscopy, PCR and sequence analysis. We named the strain as HRB isolate. The El fragment of HRB isolate, where the neutralizing epitop lies in, was amplified by PCR and determined by sequencing. The I-IRB comparied sequece isolate with other strains, we found that HRB isolate was on the same brench with the isolate strain in the 60抯, other than the same brench with vaccine strain BRD II and 2 Hong Kong isolate strain. The results suggest that the condition was probably caused by planal immunization which has not brought through by now in China and brought selective pressure on epidemic strains. The HRB isolate might provide experimental foundation for further study on vaccine which has more effective protection on epidemic strain.
