| BmDNV-5(shindai strain) was a newly isolated variety virus from silkworm (bombyx mori).At first ,the infection and proliferation of the virus in the midgut cells of silkworm were investgated by QC-PCR. The results showed that the infection and proliferation in posterior portion of midgut were more than that of oters after the silkworm was infected 3 hours and than its infection and proliferation in anterior and middle portion of midgut were increasedThen ,the complete sequence of BmDNV-5 was analysized Sequence analysis showed that the complete sequence contained four ORFs, in which ORF1 encodes structure protein and ORE nonstructure protein ;the two terminals of genmic DNA of the virus existed 232bp ITR and 161bp hairpin structure .It was similar to other insect DNV .The evolution analysis suggest that there was very higher homologous between BmDNV-5 and BmDNV-1 ,and the phylogenetic of insect insect DNV was related with die host taxonomy.Finally, we tried to construct the full-size clone of BmDNV-5 by the method of flagmented cloning then ligation. Flagmented clone were successfully constructed by sequencing, but h'gation wasn't succeeded .the two terminal ITR structure may affected the stability of the full-size clone.
