Cyanine dyes possess many good characteristics such as high extinction coefficien t, proper fluorescence quantum yied,simple routs of syntheses and steps of purification,etc. Because of excellent performance, they were used inbiological yields.The synthesis of water-soluble indole nucleus styryl hemicyanine dyes are as follow:first2,3,3-trimethyl-3H-indole-5-sulfonic acid was synthesized by using phenyl hydrazine sulfonic acid and 3- methyl- 2- butanone as starting material, and the potassium salt was obtained by reacting with potassium hydroxide, and three kinds substituted quaternary ammonium salt of N-alkyl reagents intermediates heterocyclic dyes were obtained from reacting with alkylation reagents. And then 5 benzene vinyl hemicyanine dyes were obtained condensation of the intermediate and p-chlorobenzaldehyde, p-methoxybenzaldehyde and two dimethylamino benzaldehyde. dyes were re crystallized. The structures were confirmed by 1H NMR, IR and MS, all data means that synthesized compounds are target products.Five styryl hemicyanine dyes have been designed and synthesized for improving their limited photo-stability and low water-solubility. In the synthetic process, it could be found that the sequence of the reation activity of benzaldehyde was p-chlorobenzal dehyde > p-methoxybenzaldehyde > p-dimethylaminobenzaldehyde.Thespectral properties of dyes were study, The results show the introduction of di methylamino group can improve photo stablity of the dyes, and increase the maximum absorption wavelength and maximum emission wavelength. Stokes shift is 100 nm wh en Cl were introduction to styrene ting.Styryl hemicyanine dyes that have high Photostability were study.The results show the fluorescence intensity of dyes enhance 2 times after association with BSA. The fluorescence intensity of dyes enhance 10 times after association with BSA in strong acid.All these provide a reference for biological applications of dyes.
