Squalene mainly comes from deep-sea shark liver oil, it has strong antioxidant capacity and biological activity and can be widely used in medicine, health products and cosmetics. In recent years, the study of extracting squalene from vegetable oil gradually becomes a hot spot for the invoke of protecting wild marine animals.Through the integrated process of extracting unsaponifiable matters, molecular distillation, crystallization, solvent extraction,not only high purity of the squalene was obtained from soybean oil deodorizer distillate, but also other two kinds of natural active substances: vitamin E and phytosterols in the raw materials were recoveredGC/MS was used to confirm the presence of squalene in SODD,then the HPLC was introduced to determine the content of squalene which can make sure the follow-up tests go on accurately and efficiently,additionaly the physical and chemical properties SODD were surveyed to evaluate the quality of raw materials.SODD was separated into unsaponifiable and saponifiable matter by saponification and extraction process. Then orthogonal design was applied to optimize three factors which affect the saponification reaction, when reaction time was 40 min, reaction temperature was 80℃, alkali concentration was 0.8 mol/L, the content of squalene in unsaponifiable was 10.18%.Two steps of molecular distillation were introduced to further seperate and concentrate squalene from unsaponification, response surface method was used to optimize the first-stage molecular distillation conditions, when evaporation temperature of proper was 73℃, feed rate was 1.1ml/min, wiped film speed was 251 rpm, the.purity and recovery of squalene in distillation residue were 15.2% and 92% respectively; Under the conditions of wiped film speed was 200rpm, evaporation temperature was 175℃, feed rate was 4 ml/min, system pressure was 0.1 Pa, the content and yield of squalene in distillate of second stage molecular distillation were 21.01% and 95.7% respectively.The distillates of second stage molecular distillation were crystallized to recover plant sterols, when the content of squalene was 25.3% after removal of solvent; Then squalene products(51.9%) was obtained in the hexane layer and vitamin E was recovered in the methanol layer by solvent extraction. Finally, PRO/II was explored to simulate the condition of extracting squalene using continuous vacuum distillation tower, the squalene can be enriched to 98.6% according to the the simulation results...
