Raman spectrum is the study of the molecular structure, an important tool for drug testing and trace analysis and surface vibration spectral detection research. In this article, the interaction of DNA and Amoxicillin, DNA and paeonol have been researched, as well as analyzed the S-thyminyl-L-cysteine with surface-enhanced Raman spectroscopy (SERS) techniques. The main contents are as follows:(1) The paper analyzed synthesis and post-processing method of the Ag nanoparticleso The results show that the Ag nanoparticles has the stronger SERS activity, can detect benzoic acid more and more strong enhancement signal.(2) The paper analyzed the FT-Raman spectra and SERS of the Ampicillin. The interaction of Ampicillin with Ag nanoparticles was adsorpted by O, N, S, and Ph ring. The interaction was mainly caused by the inserting-action mode between the ring of Ampicillin and the double helix structure of DNA, and Ampicillin could interact with DNA directly.(3) The paper analyzed the FT-Raman spectra and SERS of the paeonol. The interaction of paeonol with Ag nanoparticles was adsorpted by O, C=O, OH, and Ph ring. The interaction was mainly caused by the inserting-action mode between the ring of paeonol and the double helix structure of DNA, and paeonol could interact with DNA directly.(4) The paper analyzed the FT-infrared spectra, FT-Raman spectra of the S-thyminyl-L-cysteine. The optimal concentration of the S-thyminyl-L-cysteine on silver sol is 10-4mol/L. The interaction of S-thyminyl-L-cysteine with Ag nanoparticles was adsorpted by COO-, NH3+, S, and ring in the tilted way. Amino peak was enhanced in the acidic condition, carboxyl peak was enhanced in alkaline conditions. Adsorption of S-thyminyl-L-cysteine on silver nanoparticles was mainly with carboxyl under the acidic condition and with amino under the alkaline condition. Other groups were no change in different pH.
