Especially in recent years, with toxicities and side-effects of chemical medicine continuing discovered, many countries gradually aim at research of traditional herbal medicine. The basic process of studying herbal medicine contains abstraction and separation of certain active component, determination of some efficient components and corresponding preparations. With the purpose of developing dianjiang peony, we did extensive research on paeonol which is one of the important components in peony, including its abstraction, separation and preparation. The results were as follows: (1) With respect to extractionPaeonol extracted by using steam distillation and ethanol refluxing method. Comparing these two methods we found the cost of steam distillation method is lower. Therefore, the experimental conclusion of extraction of paeonol was: the steam distillation method was better than the ethanol refluxing method; the technics of steam distillation method was: the quotient of immersion solution/medicinal powder was 11: 1 ; immersion temperature was 40 ℃; immersion time was 120mins; immersion ph=6; medicinal powder granularity was about 100 screen mesh. Certain mount of sodium chloride added when steam distilling was performing will accelerate the speed of paeonol distillation but did not influence the abstraction rate. (2) With respect to preparation1) Study on paeonol inclusion complex: the inclusion technics of paeonol was determined by applying orthogonal experimental design; Qualification and quantification of paeonol-β-Cyclodextrins complex was determined by using DSC and X-ray diffraction analysis; stable constant of Paeonol-β-Cyclodextrins complex was determined by using UV spectroscopic method; compared the stabilities of paeonol-β-Cyclodextrins complex and paeonol; HPLC method was used to determine the lipid/water partition coefficient. The results were: Paeonol-β-Cyclodextrins was a new substance which is different from paeonol and cyclodextrins, Paeonol- P-Cyclodextrins content was about 87% in the ultrasonic prepared inclusion complex; drug content of was about 11%; the stability of Paeonol-β-Cyclodextrinsis was significantly better than paeonol; the lipid/water partition coefficient was significantly higher than corresponding paeonol.2) Study on preparation of paeonol troche: Using orthogonal experimental design, took the disintegration as indicator to fllterate the optimum prescription of Paeonol troche. Taking paeonol and paeonol inclusion complex as main components respectively, we prepared two kinds of troches according to the filterated prescription: paeonol routine troche and paeonol inclusion troche, further, we did relative pharmaceutical analysis of these two kinds of troches.
