Lots of antibiotics used in prevention and treatment of cow diseases,in recent years. And because the penicillin is cheap, so that was widely used by people. It is very impotent that prevents and disposes the polluted milk. This experiment chose two methods to resolved penicillin in milk. Through experiment, the effect of degradation degree to antibiotics in milk was studied. At last, to achieve the result of degradation of antibiotics.(1)Under different temperature, pH ,Ultrasonic power and time, Resolved the Penicillin in milk by Ultrasonic technology, and quantitated the residual penicillin by PHLC. The Chromatographic conditions: Phosphoric acid solution ( pH3.5±0.05 )—Acetonitrile 75:25,v/v;flow rate:1mL/min;Fluorescence detector : excitation wavelength 334nm,emission wavelength 422nm. Finally, got the best dispose condition were 25℃, pH7, 150w, 35minutes.At the moment, the residual quantity less than 1μg/L. and accord with the relevant national standards;(2)Researching the synergistic action that ultrasonic with the VC or H2O2.When the concentration of the Vc. greater than 0.2%,or the concentration of the H2O2 was 2%,then the synergistic action was obvious.But,the synergistic action was not evident when 25℃,150W, 35min, pH7;(3)Use beta-lactamase for degradation antibiotic in milk, In different pH, temperature, duration, enzyme concentration. Detecting the residual antibiotics in milk by PHLC, the Chromatographic conditions as with the (1). Experiments show that when the concentration of the beta-lactamase was 2.58IU/L,temperature was 23℃, or the concentration of the beta-lactamase was 0.22IU/L, temperature was 4℃, after 12~24h,the PenicillinG resolved completely;(4) Comparing ultrasonic with beta-lactamase, we know that the two experiments′condition within easy reach, secure and easily operated. Compared to beta-lactamase way , the ultrasonic way is cost lower; Compared to ultrasonic way, resolved penicillin more marked by beta-lactamase way.
