For the preparation of the high Enhancement ability, quality, consistent and orderly rules of SERS active substrate has become a bottleneck which has restricted the development and application of SERS, a kind of quality control, rules, orderly, easy to operate and able to attain mass production of the production process was studied. It has resolved the problem that the current SERS substrates are all poor stability and uniformity, and provided a reliable experimental data for the study of SERS enhancement mechanism.This article systematically studied the preparation and testing methods of SERS active substrate from two aspects.one is how to regulate orderly preparation of alumina pore structure of AAO template, to solve the problem of how to control the hole size, hole depth and hole pitch problems during this technology and realize the combination of sputtering and substrate isolation technology in AAO alumina template. On the other hand is based on the testing methods of surface enhanced Raman scattering.A kind of best testing method which seeking a way to play the largest SERS enhancement and responding the enhancement capacity actually was researched after studied the influence of substrate factor and enhancement factor for the SERS enhancement, so as to provides a theoretical basis for the research of SERS enhancement mechanism.Finally,we concluded that the pore-like structure and testing method had a enhancement effect on the SERS substrate.The main results are as follows:1.Using two-step anodizing method, the AAO templates with uniform pore size ,arranged in order and high pore density were prepared and studied the factors that affect the AAO template surface quality, at last reached a conclusion that surface preparation and PH value of the electrolyte solution have an effect on template quality .2. This paper has gained rules, orderly, and quality control of the SERS active substrate by the combination of AAO template with sputtering process and removing the template after the silicon substrate adhesion method. The surface structure of the substrate is ordered hexagonal close- packing arrangement, the features of which is simple process, orderly arranged, controlled structure, the consistency of the surface quality and strong empowerment.3. There is a systematic analysis of the relationship between the enhancement factor and sample concentration firstly. Test and analysis obtained: there is a big impact when Select the appropriate sample concentration and the appropriate wavelength of laser light sources, enhance the ability of the test substrate, which will has a guiding role on the testing the future of SERS substrate. |
