Objectives The activity of noble metal/titanium nitride as a Surface Enhanced Raman Spectroscopy(SERS)substrate was explored, and the drug was analyzed, which could provide reference for the application of SERS in drug analysis. Methods First, the Au nanoparticles were deposited on the surface of TiN film by electrochemical deposition method. The TiN-Au substrate showed high SERS activity.Using the TiN-Au substrate as the SERS substrate, and aspirin, salicylic acid, itraconazole, terbinafine, glucose and nicotinic acid were detected and analyzed by laser confocal Raman spectrometer. The silver nanoparticles were also assembled on the surface of TiNAu substrate to obtain TiN-Au-Ag composite SERS substrate. The existence of nicotinic acid under different p H values was analyzed, and the illegal addition of nicotinic acid was simulated by Raman detection using the TiN-Au-Ag substrate. Finally,in order to further improve the Raman activity of the TiN-Au-Ag composite substrate, the substrate was modified with N-dodecyl mercaptanon and used for the Raman detection of enrofloxacin injection. Results When the electrochemical deposition voltage and time was 10 V and 5 min, respectively, the TiN-Au substrate prepared by the electrochemical deposition method displayed strong SERS effect, and the lowest detection concentration of rhodamine 6G was10-10 mol/L.The relative standard deviation was 4.64%,and the TiN-Au substrate had high sensitivity and good uniformity. It was found that TiN-Au substrate had a certain Raman activity on aspirin, salicylic acid, itraconazole, terbinafine, glucose and nicotinic acid, indicating it could be used for the Raman spectrum detection of drugs. The TiN-Au-Ag composite SERS substrate was prepared by assembling silver nanoparticles on the TiN-Au film, and then was performed on Raman detection of nicotinic acid.In the acidic environment, the protonation of the nicotinic acid N atom mainly exists in the form of cation N+H.In the alkaline environment, the anion COO-group is vertically adsorbed on the TiN-Au-Ag surface.Imitated nicotinic acid in gypenoside solution was illegally added, and the minimum detection concentration of Raman was 10-5 mol/L.When silver nanoparticles were assembled on the TiN-Au substrate modificated by N-dodecyl mercaptanon, the concentration of N-dodecyl mercaptanon was 10-3 mol/L,the substrate had a strong Raman signal for enrofloxacin injection. The lowest detection concentration was 10-5 mol/L.Conclusions The SERS activity of the TiN-Au substrate is related to the time and voltage of the electrochemical deposition.TiN-Au substrate can be used for Raman spectroscopy detection of aspirin, salicylic acid, itraconazole, terbinafine, glucose and nicotinic acid.In contrast, the TiN-Au-Ag composite substrate has higher SERS activity, and it can also increase the Raman signal intensity of enrofloxacin injection after modification by N-dodecyl mercaptanon.Figure54;Table10;Reference 84...
