Study On Extraction And Purification Of Polyphenols From Chinese Chestnut Episperm And Its Characteristics | | Posted on:2012-05-04 | Degree:Master | Type:Thesis | | Country:China | Candidate:D D Shi | Full Text:PDF | | GTID:2131330335474662 | Subject:Food Science | | Abstract/Summary: | PDF Full Text Request | | In order to make full use of Chinese chestnut,polyphenols was extraeted from chestnut epispern and the optimum extracting method and conditions were comfirmed through single factor and orthogonal test。The antioxidant and inhibition ratio toα-amylase of the crude extract were also investigated.Polyamide and Sephadex LH-20 column were employed to separate and fractionate the crude extract and the structures of the main compounds were analyzed by HPLC一MS一MS。The main results were shown as follows:1.For ultrasonic-assisted extraction method,the optimum condition was as follows:35%ethanol as extracting solvent with a solid/solvent ratio of 1:22,extracting for 90 min at 65℃.Under this condition, the extracting yield was 19.3%, and the crude extract have the broad endurance of the temperature(<80℃) and pH(3-10).2. The antioxidant of the crude extract were studied.The results showed that when the polyphenols concentration is 10μg/ml ,the rate of scavenging DPPH was 57.42% the inhibitory rate to O2-·of polyphenols whose concentration is 100μg/ml were 47.59%; the inhibitory rate to·OH of polyphenols whose concentration is 500μg/ml were 47.65%.3. The inhibition ratio toα-amylase were also studied.The results showed that the crude polyphenols from Chinese chestnut epispern is mixture of competive and noncompetitive which belongs to reversible inhibiter, inhibitory rate toα-amylase of polyphenols whose concentration is 1mg/ml were 15.24%4. To analyse the main compounds of the crude polyphenols from chestnut epispern.5.
