| In recent years,because of the unique biological activity,the plant polyphenols is widely used in food,medicine,health products,cosmetics and other fields,have become the hotspot of study.But there are many different kinds of plant polyphenols and their structure is complex,artificial is very difficult,just can be extracted from natural substances.In order to study the adsorption of heavy metals by plant polyphenols and make full use of abundant acorn resources in our country,this experiment use acorns as raw material,study the acorn polyphenols extraction process and using ultrafiltration membrane to separation and purification the extraction,analysis the purification by high performance liquid chromatography(HPLC)method,finaly Measurement the acorn polyphenols adsorption capacity of heavy metals.The main test and the results were as follows:First,This article extract polyphenols from the oak acorns by ultrasonic and microwave assisted,compare the advantages and disadvantages of the two methods according to the extraction results,and obtain the best extraction technology conditions,extraction agent concentration of 60%ethanol,extracting time is 20 min,ultrasonic power is 300w,and solid-liquid ratio in 1:25,measured the acorn polyphenol content is 12.01%.The best technology of microwave assisted extraction:extraction agent concentration of 60%ethanol,extracting time of 21.8min,temperature of 50℃,liquid ratio of 1:25,eventually extraction yield of 13.94%.Microwave extraction method is superior to the ultrasonic extraction method.Through static adsorption and desorption experiment,for separation and purification to the preliminary separation of oak acorns polyphenols extract with the ultrafiltration membrane.To explore the optimum adsorption conditions and measuring the concentration of acorn polyphenols purified content.Using high performance liquid chromatography(HPLC)method for the acorn polyphenols composition on qualitative analysis,the results show that the acorn polyphenols sample contains acid,tannic and quercetin,also may contain flavonoids.Acorn polyphenols of heavy metal adsorption experiment results show that the acorn polyphenols of Cr6+,Pb2+,Cu2+,Zn2+ adsorption rate is very fast,within 1h,more than 70%of the adsorption of heavy metal ions,various heavy metal ions can be reached within 4h adsorption equilibrium.With isothermal adsorption model,the study found the acorn polyphenols adsorption for Cr6+ and Zn2+ can better described by Langmuir isotherm adsorption model,correlation coefficient of 0.9415,0.9924,respectively,for the adsorption of Pb2+ and Cu2+ with Frendlich more accord with the model of the isothermal adsorption.Can be concluded that the acorn polyphenols from isothermal adsorption model of heavy metal ions ability is very strong,application prospect is widespread,for further research to remove heavy metal pollution in food laid a foundation. |
