Camellia chrysantha (Hu) Tuyama is a kind of plant which in camellia genus. The former results suggested, Camellia chrysantha (Hu) Tuyama has many physiological functions such as depressing fat, sugar and cholesterol in blood serum, restraining tumor, elevating organism immunity and so on. But structure-activity relationship of saponin from Camellia chrysantha (Hu) Tuyama is not researched clearly. In this research, the saponin of Camellia chrysantha (Hu) Tuyama was extracted, and then screening the separation and purification resin for the purification of saponins. Based on the oxidation index, this research will chose the saponins components which had highest activity against oxidation for further purification, analyze its physical and chemical properties, providing theoretical basis for the structural-activity relationships of saponins from Camellia chrysantha (Hu) Tuyama.The main contents and conclusions were listed as followed:1.After the enzymes of the leaves of Camellia chrysantha (Hu) Tuyama was inactivated by microwave-heating, the concentrated solution of Camellia chrysantha (Hu) Tuyama was gained through breaking tissue, ultrasonic extraction and nanometer ultra filtration. Through ethanol precipitation and ether decoloring, the extraction solution of total saponins was deserved. Results showed that the extraction solution of total saponins mainly contain saponins and a small amount of tea polyphenols, and the content of saponins is 9 times more than tea polyphenols.2. By comparing the rate of static adsorption and desorption of the polyamide resin, XAD16 , DM130 , XAD1600 resins , the article choosing XAD16 as separate resin for the separation and purification of total saponins. The isolating results showed that total saponins was separated into WS, IS1, IS2, IS3 at the elution position of water, 10%, 20%, 30% isopropanol eluent respectively. The yield of each saponins components were 34.8%, 9.1%, 20.3% and 31.8% orderly, the recovery rate of total saponins were 96.0%. HPLC analysis results showed that, compared with the chromatogram of total saponins, the number of absorption peaks all saponins components lessened significantly, and the column chromatography of XAD16 macroporous adsorption resin can isolate total saponins of Camellia chrysantha (Hu) Tuyama into different-polarity saponins components effectively.3. In order to find out highest antioxidative saponin component from WS, IS1, IS2, IS3, taking vitamin C for reference, removing hydroxyl radicals experiment , scavenging superoxide anion experiment, scavenging DPPH·radical test and the phosphomolybdenum complex method 4 different antioxidant means were done. The results showed that each saponin component has evident antioxidant activity. In removing hydroxyl radicals experiments, the clearance for IS1 is 66.5%, higher than vitamin C and other components; In scavenging DPPH·radical test, the clearance of IS1 is 96.3%, close to vitamin C whose clearance is 100%. In scavenging superoxide anion experiment, the clearance of IS1 reaches to 64.7%, higher than reaches IS2 and IS3. In the phosphomolybdenum complex method, the antioxidant activity of IS1 is higher than IS2 and IS3. In a word,the antioxidant activity of IS1 is evident.4.The IS1A saponin component was deserved from the highest antioxidation activity IS1 saponin components by further purification through the XAD1600 column chromatography and polyamide column chromatography. HPLC analysis results showed that IS1A compose of single compound. The antioxidant activity results showed that the antioxidant activity of IS1A is 10% more than IS1, so the main effect factor of IS1 is IS1A. the results of thin-layer chromatography for testing monosaccharide species showed that the monose of saponin IS1A consist mainly of glucose and lactose. The further research for the chemical structure of sapogenin of saponin IS1A is required.
