In this study, 26 species ionic liquids (ILs) were synthesized and characterized by FT-IR and NMR techniques. It was found that ionic liquid 1-ethyl-3-methylimidazolium acetate ([Emim]OAc) was of good solubility to cellulose. The effects such as the concentration, the reaction temperature and reaction time on properties of cellulose dissolution were discussed in the dissolved system. Dissolution and regeneration of cellulose in the [Emim]OAc were investigated by FT-IR, X-ray and SEM, and the theory of cellulose dissolution in ionic liquid was explored and explained.[Emim]OAc was chosen as an environment-friendly solvent for enzymatic in situ saccharification in view of its biocompatibility with both cellulose solubility and enzyme activity. The enzymatic degradation of microcrystalline cellulose in the homogeneous system of [Emim]OAc and cellulose was investigated. Three kinds of cellulase had been selected as catalysts in the situ saccharification. The dinitrosalicylic acid (DNS) method was used to monitor the influence of reaction conditions on the results of degradation. The same method was applied to study the homogeneous enzymatic hydrolysis in situ saccharification of natural cellulose such as willow, cornstalk and pledget, in [Emim]OAc. The optimum condition for degradation was obtained in normal and IL conditions. The enzymatic hydrolysis of cellulose in ionic liquid system was compared with general conventional enzymatic hydrolysis of cellulose. And the law of the reaction was summarized. Finally, the recycling and reuse of the ionic liquid were discussed.
