| Dry roots of Uncaria rhynchophylla (Miq.) Jacks is a plant of Uncaria which belongs to Rubiaceae. It distributes in Yunnan, Guangxi, Guangdong etc.The pharmacology research indicated that the plant of gouteng have the properties of heat-clearing, blood circulation promotin, expelling wind, dilation of blood vessels, regulation of the immune. Alkaloids, saponins, flavones, coumarins, terrenes, esters, fatty acids, enolase, olefins and phenolic compounds,The medicinal plants often have great difference in effective constituents owing to their producing areas and growning periods.But the study of chemical components and Pharmacological of Guanxi Gouteng is deficient. So, The chemical components of Gouteng has been furtherly studied in this thesis,it is very useful for development resource and provide certain scientific basis.The antioxidant activity of petroleum ether extract (PE), chloroform extract (CE), ethyl acetate extract(AE),ethanolic extract(EE), alkaloids(Alk) of Uncaria rhynchophylla (Miq.) Jacks were determined with Flow-injection chemiluminescence and compared to Vitamin C (Vc). Their IC50 value were 20.432 mg/mL,1.547 mg/mL,0.0283mg/mL,0.00326 mg/mL,15.298 mg/mL, IC50 value of Vc is 0.0223mg/mL, So the results showed their antioxidant activities are in the sequence of EE>VC>AE>CE>Alk>PE.The 75% ethanolic extract of gouteng, total alkaloids was obtained from the gouteng by acid extraction and alkine precipitation, and solica gel for several times, two alksloids were obtained and identified as Isorhynchophyline (6), Rhynchophyline (7). Meantime, the 95% ethanolic extract of the crushed gouteng was re-extracted with petroleum ether, chloroform, ethyl acetate successively to get three fractions.Fourteen compounds were isolated from the chloroform extract by column chromatography and nine of them have been defined by spectroscopic methods as follows: Hyperin(1), Physcion(2), Vanillic acid(3),β-sitosterol(4), hydroquinone(5), Stigmasterol(8), Quercetin (9),Oleanicacid(10),Daucosterol(11). compounds 1,5,9 were first isolated from Uncaria rhynchophylla (Miq.) Jacks.L9 (34) orthogonal test of Fatty acids preferred supercritical fluid CO2 extraction (SFE-CO2) process conditions, the effects of pressure, temperature, time, and CO2 rate of flow for the study of factors, and the yield of Fatty acids for the evaluation of indicators. The Fatty acids optimum extraction conditions by SFE-CO2 are as follows:pressure 40MPa, temperature 30℃, time 3 h, and 30 L/h CO2 rate of flow.And its composition was separated and identified by GC/MS analysis. |
