| High performance liquid chromatography (HPLC) has been successfully applied to extract the biologically active compounds from natural resources. The effective components in hop extracts was seperated and purified by HPLC in this paper, and the structures of final products were characterized and verified in the end.Firstly,6 kinds of active ingredients in hop extracts (humulone, cohumulone, adhumulone, lupulone, colupulone, adlupulone) were baseline separated by the analytical HPLC in order to scale-up and purity characterization.Secondly, the preparative chromatography conditions were determined after the linear magnifying technology. Moreover, the effect of mass and volumn overload was also investigated to obtain the optimal chromatographic conditions:colomn (Hypersil C18 lOμm), room temperature, mobile phase V(CH3CH2OH)/V(0.2%HAc)=65/35, flow rate 15.0mL/min, injected capacity 90 mg, detective wavelength 315nm. The eluted solution obtained was evaporated under vacuum and extracted by SPE, then dried after the nitrogen blowing to get the yellow solid products.Finally, the structures of final products were identified by UV, FT-IR and LC-MS. The results showed that the purity of products was more than 90% by analytical HPLC technology.The preparation method in this thesis is simple and has a high purity. As a result, the application by this way will guide its industrialization and develop applied methods in the near future. |
