Works including the extraction of alkaloids derived form the hull of water chestnut have been reported in this paper first. The alkaloids are separated by the high performance preparative liquid chromatography after purified. Three compounds are got and their constructions are determined. 1. The best conditions and methods of extracting alkaloids are identified through comparing the effects of different solvents and methods such as acid solution constant temperature water-bath extraction, acid solution ultrasonic extraction, 95% ethanol constant temperature water-bath extraction and 80% acid ethanol constant temperature water-bath extraction. 2. The alkaloids are separated and purified by polarity-grads solvents and macroporous resin. The adsorption efficiency and disadsorption efficiency are investigated together with the effect of adsorption time, adsorption temperature, kind of macroporous resin, etc. It is the best to use AB-8 macroporous resin and 80% acid ethanol, which is the elution solution, to purified the crude sample of alkaloids. 3. The best expanding solvent system used in the analysis of thin-layer chromatography is selected through comparing the separation of different solvent systems with different polarity. The results not only confirm the purification by macroporous resin but also can be used to select the composition of mobile phase in the separation and preparation by chromatography. 4. The best separation and preparation conditions of high performance preparative liquid chromatography are selected and identified through comparing the composing of mobile phase, flow speed and so on. Three components have been separated and the basic structure of one alkaloid is shown after IR, MR, 1HNMRand MS analyses.
