But, we still haven't find the appropriate material and the shaping technics.As a result, the study choose the appropriate material, whose degradation time match with the growth time of the cell planted on it. It means, the material of the scaffold will be totally degraded when the cell grows a new urethra. In this situation, the urethra won't be blocked and won't cause complications.The demand of the shaping technics is the imitating of the urethral appearance, and the hole ratio is above 85%, which not only can keep the nutriment pass inside and outside, but also keep the cell from losing in the hole. Busides, the urethral scaffold has a good mechanical property, included tensile force, twist and radial compress. Finally, the study choose the material PGA, using braided art to carry out the biological and mechanical requirement of the urethra.In the planting, the key is to promote the adhension between the cell and scaffold, which can improve the cell livability. The study chose the low temperature plasma and RGD inarch to change the surface character which can improve the adhension. The rule is not to distort the scaffold and not to reduce the mechanical property. Especially in the degradation period, the scaffold will be break and dent which will cause a lot complications. The study choose the appropriate kind, temperature and time of plasma, aim to improve the biocompatibility and avoid the decreasing of mechanical property.The animal test is to exam the urethral scaffold invent in the study. Plant the bladder mucosa cell on the scaffold, observe 1 week in the bring up box, then transport to the mice skin 2 weeks imitate the situation in body. At last, replace the urethra of the buck in no bacteria infection. In the test, use the HE coloration and X-image test to observe the urethra in the buck, whether the cell layer be thicker, the scaffold be degradated, the urethra be smooth.The study is the first urethral scaffold by braided art, which can imitate the shape and biological and mechnical property. Use the low temperature plasma and RGD inarch to improve the adhension between scaffold and cell, plant the bladder mucosa cell and then transport to buck. It can replace the urethra, and it will be a new urethra when the PGA totally degradated and won't cause any complications. |
