| In the peel removing and gum extracting experiment,a large number of single-factor experiments and orthogonal test was used to access the interactional condition and the optimized processing parameters.The theology experiments showed that the apparent viscosity of the fenugreek gum Exponential increases with the concentration of the solution. The solution was non-Newtonian and it has instant viscosity-recovery property when it was treated with shear thinning.Its viscosity decreased when the temperature was increased or if the solution was treated with freeze thawing.Fenugreek gum solution was stable in alkaline conditions.Fenugreek gum had strong synergistic effect with Carboxymethylcellulose(CMC) and a bad one with xanthan gum or sodium alginate.In the sulfation experiment of fenugreek gum,the Wolfrom method and concentrated sulphuric acid method were used to prepare sulfated fenugreek gum.The optimal technical parameters in Wolfrom method were:fenugreek 7g,the ratio of chlorosulfonic acid and pyridine was 0.3,under the conditions of temperature 30℃reaction 2h,the subsitituion value was 0.92 and the viscosity of the solution was 137.1mpa.s;The optimal parameters of the concentrated sulphuric method was 7g of fenugreek gum,0℃,3mL of concentrated sulphuric acid and 6mL of butanol,the substitution value was 0.3 and the viscosity of the solution was 228.3 mpa.s.After esterified,the solution of the sulfate fenugreek gum which was prepared by the Wolfrom method showed a better substitution value and the the solution of the sulfate fengureek gum which was prepared by the concentrated sulphuric method showed a better viscosity than the former.In the phosphorylation experiment of fenugreek gum,the phosphorus oxychloride method and the semidry process method were used to prepare phosphorylation fenugreek gum.The optimal technical parameters in phosphorus oxychloride method were:7g of fenugreek,3mL of phosphorus oxychloride,reacted under 0℃for 1 hour,the substitution value was 0.92 and the viscosity of the solution was 197.6mpa.s;The optimal technical parameters in semidry process method were:10%of Mixed phosphate,5%of carbamide, reacted under 70℃for 1 hour,the substitution value was 0.026 and the viscosity of the solution was 246.9mpa.s;After esterified,the solution of the phosphorylate fengureek gum which was prepared by the phosphorus oxychloride method showed a better substitution value and the solution of the sulfate fengureek gum which was prepared by the semidry processing method showed a better viscosity than the former.After esterified,the molecular size of the fenugreek gum reduced.And the viscosity was lower than before.They declined with the increasing of the substitution value.The sulfated fenugreek demonstrated a better solubility and Transparency while the phosphorylate fenugreek gum showed a better viscosity.The sulfate fenugreek gum and the phosphorylate fenugreek gum had some degree of antitumor activities.The result showed that the suppression ratio of sulfate fenugreek gum(800μg/mL,0.87 DS) on S180 and H22 were 81.13%and 59.02%seperately,the suppression ratio of phosphorylate fenugreek gum(800μg/mL,0.87 DS) on S180 and H22 were 58.09%and 46.12%seperately. Compare the viscosities,solubility and luminousness of the four esterified fenugreek gum:sulfate fenugreek gum,phosphorylate fenugreek gum,carboxy methylation fenugreek gum and hydroxypropylation fenugreek gum;the result showed that phosphorylate fenugreek gum had the biggest viscosity while the carboxy methylate fenugreek gum had the biggest solubility and luminousness. |
