Constrast To Separate Gingerols By AB8 Macroporous Resin And Silica Gel Column | | Posted on:2010-04-23 | Degree:Master | Type:Thesis | | Country:China | Candidate:C Y Gu | Full Text:PDF | | GTID:2121360278972823 | Subject:Chemical Engineering | | Abstract/Summary: | PDF Full Text Request | | Ginger oleoresin was extracted by solvent extraction.The separation system was chose on base of the structure and the nature of isolated components gingerols.The effect of separation factors was researched in the the process of experiment.First of all,gingerols standard sample purchased from Sigma company was analyzed through high-performance liquid chromatography(HPLC) and mass spectrometry(MS),gas chromatography-mass spectrometry(GC-MS),ultraviolet(UV) spectrum.HPLC and MS analysis showed that the molecular mass of the highest content composition(98.5%) in the gingerols standard sample was 294.2.GC-MS analysis results proved that gingerols can transfer into zingiberone.UV spectrum was chosen to analyze the content of samples in the process of experiment.UV spectrum analysis results showed that concentration and absorbence intensity of ginerols was linear and its linear equation between concentration and absorbence intensity wasY=0.001540+0.01186X(R=0.99998)Second,ginger oleoresin was extracted by solvent and analyzed by GC-MS.The results showed the peak area percentage of gingerols was 1.27%and 9.22%when the peak area percentage was calculated including zingiberone.Third,ginger oleoresin was separated by silica gel column chromatography.UV spectrum and GC-MS was used to analyze the samples.The UV spectrum results showed that the gingerols mostly in the paragraph 3 and 4 in which the highest part was in the paragraph 3. GC-MS results showed that the relative content of gingerols was 9.55%in the paragraph 3 and 14.34%in the paragraph 4,when zingiberone was included,the relative content of gingerols was raised to 17.61%in the paragraph 3 and 15.04%in the paragraph 4.The recovery of the paragraph 4 was 59.01%.Finally,AB8 macroporous resin was selected as the separation system based on the structure and nature of gingerols and macroporous resin.In the experiment we researched different conditions to separate the active ingredient gingerols in ginger oleoresin.The content of gingerols in the samples was analyzed by ultraviolet spectrum and GC-MS.Then the standard gingerols sample was used as reference to determine the recovery of gingerols. The results showed that the ether and hexane-eluting agent system can bring an evident result. On the one hand,the kind of components reduced from 27 to 11.On the other hand,the peak area percentage of gingerols raised from 1.27%to 55.68%.The recovery can be up to 70.88 %.Contrasted to silica gel column and AB8 macroporous adsorption resin,It should be concluded that latter method was better than the former method to separate gingerols in the ginger oleoresin.The amount of ingredients in the paragraph 4 separated by silica gel column chromatography was 19 and they accounted 84.83%in the gross peak area in which zingiberene was 13.34%and gingerols was 14.34%.The total of ingredients separated by AB8 macroporous adsorption resin was only 11 and they accounted 85.64%in the gross peak area in which gingerols was 55.68%. | | Keywords/Search Tags: | gingerol, AB8macroporous adsorption resin, silica gel, ginger oleoresin | PDF Full Text Request | Related items |
