Ginger(Zingiber officinale Rose)is the rhizome of the Zingiberaceae.6-gingerol?6-shogaol?protease in ginger had a wide and high application value.The main research contents and conclusions of this thesis were as follows:1.The HPLC method for detecting the main components of ginger in this laboratory had been re-established,the results showed that they were all good.It provided that this method was available.The result of detection of different ginger and products showed 6-shogaol was the product of post-processing.And the other three ingredients could be affected by multiple factors.Supercritical CO2extraction method was an effective process method to extract gingerols in ginger.2.The drying experiment was divided into single part such as single hot air drying and microwave drying method and combination drying process by hot air and microwave.In single part,a drying rate curve and a wet-base moisture content curve were established first.The results show that the hot air temperature and microwave power dry faster with the increase of conditions,and in terms of speed,the trend of first increase and then decrease was presented,It was similar to the literature.Gingerols were mainly studied for the content of 6-gingerol and its ratio to the content of 6-Shogaol.The result showed that the content and ratio were reached the peak at 60?temperature,the wind speed was at the first gear,which was the minimum wind speed,the microwave power was 150w.The combined part adopts the drying method of hot air combined with microwave.Under single factor experiment,the results showed that the best value were under 60?temperature,1 gear wind speed,30%the water content at the conversion point and 150w microwave power.The predicted temperature was at58.84?,and the water content at the conversion point was at 24.95%,the wind speed was at level 1,the predicted content of 6-gingerol was at 9.77mg/g,and the maximum ratio was at 1.31%.The results of the verification test showed that the content of 6-gingerol was at 10.23mg/g and the ratio was at 1.5%,which was close to the predicted value,and the combined drying method had certain advantages compared with the single drying method in quality.3.In the extraction of ginger protease,we established two standard curves for the detection of protease content and enzyme activity,and then investigated the material-to-liquid ratio,extraction temperature,buffer p H solution value and citric acid concentration during the extraction process.It showed that the optimum value was the1:3 material-to-liquid ratio,20?extraction temperature,6.5 buffer solution p H value,and 20mmol/l citric acid concentration,and then the protease content was at 0.65%,the protease activity was at 463.75U.Subsequent purification which used by dialysis bag showed that the result of ginger protease content was at 0.80%,the protease activity reached at 440.435U,and the clear liquid can still detected 13.95?g/ml ginger protease.Enzyme specific activity is 1.03 times of that unpurified.Based on the date above,it confirmed that this step had a certain purification effect.
