| 1 BackgroundThe progress of wound healing includes stoping bleeding in earlier period, protecting wound surface,contoling inflammation,histiooytic hyperplasy,forming granulation tissue,creeping of the wound limbus,and so on.Proliferation of endothelial cell,epithelial cell,and fibroblast is the key point of forming granulation tissue and wound healing.Presently,the drugs for external use which can promotes wound healing include:(1) Filling wound cream,which can fill the wound and contol inflammation.(2) Fu kang ning,which can enhance organism to generate more immunocell and help to move the cell to the wound.(3) Mist spray to patch wound, which can form a film to protect the wound and send active component to penetrate skin.(4) Benzalkonium chloride paster,which can sterilize,stop bleeding and protect wound.(5) Yunnan white powder and it's styptic plaster,which can help platelet aggregation,prevent tissue from swelling,and promote the expression of bEGF and VEGF.But it's main effect is to stop bleeding and prevent tissue from swelling in earlier period.It's help of promoting the proliferation of endothelial cell,epithelial cell,and fibroblast is small in midanaphase.The aim of our research is to break through in this area.Ecdysterone(B-ecdysone) is a kind of hormone of hexapod to modulate metabolic regulation.It was researched by entomologist in early time.Hexapod, scrustacean and arthropod have callous corneous layer.They secrete ecdysterone to exuviate the comeous layer regularity and grow new layer.This process is called ecdysis.But it was found that ecdysterone exists in vegetable kingdom more than in animal kingdom.The researchful application of ecdysterone is mostly depend by distill of plants by now.Ecdysterone is indispensable to vertebrate.It's main mechanism is to:(1) promote protein synthesis,(2)modulate glycometabolism,(3)modulate lipid metabolism,(4) modulate gene expression,(5) improve immune function,(6) act to central nervous system,(7) act to cardiovascular system,(8) promote concrescent.Products of ecdysterone in the market at present include:invigorator(Ecdybol and EcdyZonc which can promote protein synthesis of muscles),cosmetics (radiation-proof Feflex L-Camitine and whiten cosmetics of SUMA),thymoleptics (L-Phenylalanine,DL-Phenylalanine,SMAR),drugs(Ecdisten which can be used to treat trichomoniasis,Phytoadaptogens which is anti-arrhythmic,Iterations which is hypolipidemic,Ebselen which is anti-inflammatory,E-1110 and E-2002 which are antineoplastic agents),and so on.2 ObjectiveTo study how to manufacture cream preparation of ecdysterone with ecdysterone monomer,and use the ecdysterone cream to treat wounds in rabbits to make a dose-effect analysis of the ecdysterone cream,and deduce the optimum effective concentration.3 Methods3.1 Preparative methodMeasured a certain amount of glyceryl monostearate,octadecoic acid,petrolatum album,azone,and ethylparaben.Heated them to 90℃with bain-marie and melted to oil phase.Measured a certain amount of distilled water,borax,irium,trolamine, glycerine,and allantoin.Heated them to 90℃with bain-marie again and melted to aqueous phase.Measured different weights of ecdysterone monomer and dissolved them with alcoholic,then got different concentration of ecdysterone solution.Putted the oil phase to the aqueous phase slowly and agitated to get cream base.At the same time,putted one concentration of ecdysterone solution to the cream base.After agitating 30 minutes,putted the mixture to the atwirl emulsor with the rotation speed of 2000-6000r/min-1,then got one concentration of ecdysterone cream.Used the same method to prepare the other three concentrations of ecdysterone cream.After that,detected the quality of the four concentrations of ecdysterone cream with high efficiency liquid chromatography.3.2 Dose-effect analysis methodMade animal models of separated full-thickness skin of 11 Zelanian rabbits which had close weights.There were five wounds on the back of each rabbit.Then applied the cream base and the four concentrations of ecdysterone cream to the five wounds apartrespectively.On day 4,8 and 12 right after the models were formed,the healing area and the percentage of the healing area were computed after every wounds of 8 of the 11 rabbits being taken pictures.The local wound tissue of the rest 3 rabbits was obtained for pathological examination on day 4,8 and 12 respectively.4 Results4.1 PreparationThe ecdysterone creams were prepared successfully.The measured content of ecdysterone before each cream was prepared was closely to the content of ecdysterone cream which was prepared.The variance could be ignored.Further more, the chemical character of ecdysterone wasn't changed in the progress of the ecdysterone cream preparation.4.2 Dose-effect analysis Statistical analysis of the percentage of the healing wound area showed that the ecdysterone cream can promote wound healing(P<0.01),and its optimum effective concentration was 2.5%.Pathologically,the wounds of the 2.5%ecdysterone cream group exhibited best effect for shaping the granulation tissue and stimulating the proliferation of epithelial cell,endothelial cell and fibroblast.5 ConclusionThe ecdysterone cream can obviously promote wound healing in rabbits,which may offer a clinical alternative for promoting wound healing.Its optimum effective concentration is 2.5%. |
