| As is well known, the active oxygen in the body has close relationship with the people's consenescence. its production and clearance is at dynamic balance. Many studies indicate that the active oxygen does harm to cells evidently, for it can make protein lose activity or hydrolyzed, and break DNA off, and peroxide the lipid membrane. In such way, the structure and function of cell are damaged. nowadays, the function food which is used to eliminate active oxygen are nearly non-SOD food and SOD cosmetic, which only does good to beauty as respect to skin, and has no effect on all organs from the point of fighting consenescence. So it is neccessary to develop the high efficacy functional food with SOD.Nowadays,the SOD products mostly come from blood of animals such as cow,horse, pig, chook,dog and so on in our country,and those materials are probably contaminated the danger of virus and bacteria infection,as well as the contaminations coming from the feeding process.so the study of obtaining SOD from plant become more and more hot internationally,such as SOD extracted and purifed from garlic,leek and corn flour.The results of animal experiment in the lab indicate that the SOD from plants have the same function with that from animals,which predicts the great foreground of developing the plant SOD.Chaenomeles sinensis fruits is one of the ten-best foods recommended by the nutritionist in our country,which is regarded as not only food but also medicine publicized by the national department of health,for it has rich nutriment and active substance such as animo acid,vitamin and mineral. Previous studies indicate that it has functional and medical efficacy,such as anti-inflammation, easing pain,protecting liver, inhibiting tumour and antibacterial function.However, there are few reports of superoxide dismutase(SOD) from Chaenomeles sinensis.Through our pilot study,we find there are not only flavone and polyphenals,but also superoxide dismutase(SOD) in the Chaenomeles sinensis. Since the cyclodextrin(CDs) was obtained from 20th century, there are more and more new progress to their research constantly. Not only the output of CDs is improved, but also the structure of natural CDs is modified to a series of CDs derivative with unique performance. At present, CDs is applied to food processing, .cosmetics and chromatography etc, except for extensive uses in medical industry. The inclusion complex of medicine withβ-cyclodextrin is with better solubility, bioavailability, and stability, also with less odd flavor and irritant. In recent years, the application of CDs in the food industry is more and more extensive, especially protecting the biological activity of active substance such as polypeptide, protein, enzyme, improving their stability, lengthening half-life, and delaying delivery. To apply CDs inclusion technology on Chaenomeles processing, it is new protocol to dealing with Chaenomeles material in research, which could improve their bioavailability and stability as product.In our study, firstly, the main antioxidant substances in Chaenomeles sinensis were investigated,and mainly emphasized on the determination and purification of SOD; secondly, enzymatic process, decoration of SOD with CDs and spray-to-produce-particle technique were used for preparing SOD antioxidant particles of Chaenomeles Sinesis(CSAP), dramatically increase its extraction rate and stability for storage; thirdly, irradiation animal model was established for assessment of antioxidant activity of CSAP.The main results were:1. The assay results indicate that the total activity of SOD in the Chaenomeles sinensis is 4.32×104U/100g,almost Cu,ZnSOD accounts for 95% of total SOD activity,that is 4.10×104U/100g.2. Other antioxidant substances in Chaenomeles sinensis includes polyphenols and vitaminC,the polyphenols and vitamin C in fruit juices are 7.8mg/mland 0.65mg/ml,among the total polyphenols,tannins accouts for about 50%,and flavonols accouts for about 16%,other polyphenols are 34%.3. After the seperation and purification of SOD,the SOD purification folds, the yield and the specific activity are 32.4, 11.1% and 6159U/mg respectively.The molecular weight of SOD determined,which is between 25KD and 31KD,almostly 28KD.4. Taking the results of single factor experiment, orthogonal experiment,the optimal enzymatic parameter is abtained: the proportion of materials and water is 1:2,enzymatic pH is 3;the addission of pectinase-cellulase is 1.2 %;the enzymatic temperature is 40℃and enzymatic time is 60min.After enzymatic precedure, dramaticly increased the total SOD activity,that is 10.65% more compare to that before enzymatic procedure.5. Using 3% ofγ-dextrin to pack SOD get the better result.The packed product reserve 40%,52% and 34% SOD activity respectively,after thermal treatment(50℃,45min), acid treatment(pH=1.7,15min)and artificial gastric juice treatment(15min).6. From the result of orthogonal experiment,the ratio of material to fluid is 2:1,and the work matetial temperature is 45℃.In this way,compared with total SOD activity in the centrifugate liquid,the SOD activity reservation rate is 67.8%.There are 2.4×105U SOD activity in 100g CSAP.7. During the storage,the SOD activity,contents of total polyphenols and vitaminC seperatily decreased 62%,35% and 23% in frozen fruits,but 34%,11% and 59% in CSAP,CSAP indicats good quality for storage.8. The results of aninal biological function test indicates:the MDA content in blood serum and organizes of every group with different dose is lower than the model compared group,and the SOD activity in that is higher than the model compared group.So, the CSAP has significant contribution to reduce MDA content in blood serum and organizes of mice,while icrease the SOD activity in that. |
