Polychlorinated biphenyls is noted as an chamical organic matter with toxicy. It can affect the healthy of organism by enriched in the humen's body. It can be absorbed by organism's skin lung and stomach, when it is flowed to liver organize of muscle and fattiness by blood. The toxic effect to humans is can't be reversed. Because the traditional methods to dispose polychlorinated biphenyls all exist deficiency, this task study the effect of microorganism degrading polychlorinated biphenyls owed to these causations.Because of the chlorine atom replace the hydrogen atom which on the phenyl, this structure of polychlorinated biphenyls make it being be degradated and it is poison to microorganism. And so it is very valuable to select one of microbe strain can tolerant the toxicity of polychlorinated biphenyls and also can degrade it .This experiment choose the wastewater and the activity sludge or the soil as the source of microbe which in Songdong Wastewater Purified Company, ROTORK Control LTD, Shanghai Tianyuan Chlor-alkali factory. The biphenyl was used as alone carbon source to cultivate the microorganism. When the microorganism was domesticated first time, enriched, domesticated second time, separated and purified, it was separated three kinds of strain which can degrade biphenyl. These three kinds of strain were inoculated to the culture medium with biphenyl. After two weeks the degradation efficiency of biphenyl tested by liquid chromatography can up to 60%-70 % . Especially the microorganism called W-02 strain which was chosen from ROTORK Control LTD has the best action to biphenyl, the degrade efficiency can up to 70%.W-02 strain was studied after it was determined that it can degrade the biphenyl with the best result. When the incubation media was added the extra carbon, the growth situation of W-02 strain in it is worse than in the incubation with biphenyl as the only carbon source. In the condition of variant pH, temperature and the speed of rotation, its growth situation was tested to find the best one. So it was confirmed that the best enzymatic reaction conditions: the pH is 7.0, the temperature is 35℃when it is in the constant temperature incubator.Then it was analyzed by Gram's staining to determine the initiative character. The gene sequence and the microbe culture was tested by Shanghai GeneCore BioTechnologies Co.,Ltd. The strain is Staphylococ-cus epidermidis though the 16SrDNA-gene analysis. It belongs to Micrococcaceae and Staphylococcus.The reason why the polychlorinated biphenyls can be degraded is that the particulaly enzyme can oxidize the benzene ring.And the polychlorinated biphenyls would turn into long chain organic matter and be degraded. When the enzymes were deposited and separated by chromatography, three kinds of enzyme was separated. The enzyme marked number I has the best activity tested by UV-Visible light analysis. In the condition of the temperature is 30°C and the pH is 7.0, The enzyme has best activity. And the magnesium ion can accelerate the activity of the enzyme. The calcium ion, the ferri ion, the zinc ion and the aluminum ion can suppress the activity at a certain degree. However, the cuprum ion and the iron ion suppress the enzyme activity totally.The culture was inoculated to the watstewater and test the COD.It is show that the culture as the only microbe in wasterwater has illdefined action to COD.But when the culture was inoculated with other activity sludge,the action was obviously,the result of treatment is better than with only culture or with only activity sludge. But when times gone the result of wastewater treatment with mixture culture was approach to the wastewater with activity sludge.It is indicated that the seperated culture was assimilated by the indigene microbe.How to use the strain W-02 to treat wastewater effectively is a problem to us.
