Triazole compounds have been found to show wide spread biological activities, many of them have been developed and used as fungicides, plant growth regulators and medicine. Triazole compounds have been the most important of types of fungicides in nitrogen hencrocyclic compounds. Different structures of triazole compounds have different biological activities, developing novel structural triazole compounds is the research focus on at present.Triazole and its derivative as ligand has the advantage of small size, great ability of coordination and the multiple points of coordination, it can also coordinate to the metal ion and others ligand with many forms. It is also the good N-Heterocyclic system, its -N N- group has high absorbing electron, and its existence can make N-3 atom of triazole connect every kind of polar group awfully. So triazole compound is one of the most action field of development which studied on the synthesis in recent years. Therefore, this compound also comes under the large of chemists general attention. As the coordination form of bridging ligand will diversify in virtue of three nitrogenous atoms on the ring. Triazole and its derivative as ligand join in the synthesis of metal complexes. This research will become one of the hot research field gradually. This paper mainly studies on the synthesis of water-soluble 2-(4-N-1,2,4-triazol) propanoic acid-(4-ylimino-1,2,4- triazolc)(Trzpa); water-soluble 3, 5- 2 (4-pyridyl) -4-amino-1,2,4-triazol(G) and metal complexes, the interaction with the biological molecular DNA, in order to provide the theoretical and experimental basis for synthesis the efficient, low poisonous and water-soluble compound.This thesis is constituted by four parts:Part I : ReviewIn this thesis, it has been reviewed the recent advances in the studies on the structures, synthesis, action mechanism, structure-activity relationship and fungicidal activities of triazoles; Triazole compounds have five main type; The application of triazole compounds in pesticides and medicines; The application of complexes aspects of metal that containing nitrogen heterocyclic ring. Triazole compounds synthesis methods; The research progress of triazole and its derivativc-mctal coordination; The article introduces the basis of choice the subject, contents and significance of the research.Part II: Synthesis, characterization and properties of 2- (4-N-1,2,4-triazolc)propanoic acid-(4-ylimino-1 ,2,4- triazole) and if scomplexcs2- C4-N-l,2,4-triazol) propanoic acid-(4-ylimino-1,2,4- triazol) (Trzpa) has been synthesized by the reaction of 4-amion-l,2,4-triazole and acetone acid in alcohol solution (pH 7~8),and it's complexes have been synthesized by the reaction of metal nitrite with Trzpa in water solution (pH-5-7). The composition and thermal stability of Trzpa (C7N8H10O2) and the complexes (M2(Trzpa) (NO3)n·nII2O) were characterized by elemental analysis, molar conductivity, thermal analysis, IR, HNMR, UV. methods.Part III: Study on the interaction of Trzpa and it's Cu(II),Co (III) complexes with DNATo study the interactions of Trzpa and it's Cu(II),Co (III) complex between DNA , the Uv-Vis spectral, fluorescence spectral, viscosity measurements and CD (circular dichroism) methods were used. The results indicate that in the presence of ligaid Trzpa and the complexes, DNA exhibits a hypochromism in the u.v. vis. spectra. The emission intensity was quenched as ligaid Trzpa and the complexeswere added to the EB-DNA system. The binding constant :K£=1.1×103 L / mol(ligand Trzpa); K=1.8×104 L/mol (Cu) ; K=1.3×103 L/mol (Co) at 20℃. The negative spectra of the CD and the viscosity of DNA is changed by adding Cu (II) complex. The above results suggest that the Cu- complexes can bind to DNA mainly by intercalation, revealed by competitive experiments, viscosity and CD studies.But, the Liganid Trzpa and Co( III )complcx bind to DNA by no intercalation model. Part IV: Synthesis, characterization of 3, 5-bis (4-pyridyl)-4-amino-1.2,4-triazoland it's complexes3, 5-bis (4-pyridyl) -4-amino-1,2,4-triazol (G) has been synthesized by the reaction of Hydrazine hydrate and isonicotinic in alcohol solution (pH 7~8), and it's complexes have been synthesized by the reaction of metal nitrite with G in water solution (pH 5-7). The composition and thermal stability of G and the complexes were characterized by elemental analysis, molar conductivity, thermal analysis, IR, HNMR methods.
