Probiotic strains of lactobacillus is the warm spots in recent years, and the cholesterol-reducing ability of lactobacillus isolated from a Traditionally Home-Made Kumys catch the interest of scientist , The study of the mechanisms of the cholesterol-reducing ability of lactobacillus is not only significance in biology but also enhance the generalization of therapeutic action of kumys。In this paper,6 lactobacillus strains isolated from a Traditionally Home-Made Kumys were studied, subcultured in MRS medium,, observed and recorded there cell morphous。By the technique of PCR(polymerase chain reaction), cloned and Characterization of the bile salt hydrolase genes (bsh) lactobacillus strains isolated from a traditionally home-made kumys, obtained 940bp DNA fragment,and coding 313 amino acids, Comparison of the predicted amino acid sequence to the database by BLAST analysis revealed highest similarities to the BSH of B. longum strains (91% identity and 95% similarity). Significant similarities were also found to the BSH enzymes of several Lactobacillus strains (35 to 43% identity)(GenBank accession numbers AF054971, AF305888,AF091248, AF297873, and M96175), as well as to BSH enzymes of Enterococcus strains (36 to 37%) (GenBank accession numbers AY032999 and AY260046), L. monocytogenes (37%), C. perfringens (36%) (7), and penicillin V amidase (PVA) of Bacillus sphaericus (30%) (GenBank accession number M15660)...
