49 strains were isolated from 16 samples of traditionally home-made koumiss collected from the pastoral area in Inner Mongolia-XiLinGuoLe region. All of them belonged to Lactobacillus. According to the traditional morphological characters, physiological and biochemical properties, as well as chemical characteristics, they were identified to 10 species, namely, Lb.casei(16 strains), Lb.acidophilus Group(10 strains), Lb.plantarum(6 strains), Lb.kefiranofaciens(2 strains), Lb.curvatus (1 strains), Lb. paracasei subsp. pseudoplantarum(3 strains), Lb.coryniformis subsp.coryniformis(5 strains), Lb.fermentum(1 strains) and Weissella kandleri(1 strains), 2 strains had not been identified yet. To further differentiate Lb.casei.Zhang and ZL12-1, the genes of 16S rDNA were amplified in vitro and sequenced. Then a phylogenetic dendrogram was constructed by comparing these two sequences with other 16S rRNA/DNA sequences of Lactobacillus. The results indicated that the homology of Lb.casei.Zhang with Lb.casei ATCC 334 was 100%, and the similarity of ZL12-1 with Lb.gallinarum ATCC 33199 was 98%. Combined with analysis of the phyolgenetic tree and partial sequences of 16S rDNA , Lb.casei.Zhang was classified as Lb.casei subsp. casei, ZL12-1 belonged to Lb.gallinarum. The result was as same as that of traditional classification before. |
