A detail procedure for extracting active polysaccharides from Eucheuma striatum, a kindof big economical algae in South-China Sea, has been developed by using amicrowave-assisted extraction(MAE) device. Based on the study of our laboratory, amagnified experiment for extracting active polysaccharides from Eucheuma striatum wascarried out by the way of the hot water extraction. Finally, the biological activity test was usedand the results confirmed that both Microwave-assisted extraction and magnified experimentof the hot water extraction could protect the biological activity of polysaccharides. Thisconclusion could do good to the industrial production of the active polysaccharides.The Influential factors during the extraction process have been confirmed. The extractionof polysaccharides was influenced obviously by solid/solvent ratio, microwave power andextraction duration respectively; the polysaccharides could be complete deposited if thealcohol concentration was more than 70%. In addition; the quality of the polysaccharidestreated by the vacuum-freeze drying and reduced pressure heat-air drying was obviouslybetter than those by the normal pressure heat-air drying. The analysis of orthogonal test andthe Box-Behnken design test were carried out. Based on the analysis, effects of solid/solventratio, microwave power and extraction time were explored, the optimum extraction conditionwas obtained as follows: 560W microwave power for 12min extraction duration incombination with 75:1(volume/weight) solvent and solid weight ratio. Under the optimizedcondition, the extraction rate of polysaccharides reach 68.57%. The result showed that it'snondistinctive between the method of the response surface analysis and orthogonal test. Themicrowave-assisted extraction is more efficient than hot water extraction.A magnified experiment for extracting active polysaccharides from Eucheuma striatumwas also carried out by the way of the hot water extraction. During the extraction process,erery unit operation and the Influential factors were confirmed. The qualities of thepolysaccharides and their biological activities included. The result showed that the optimumextraction technical route was as follows: First, smashed the dry alga to granule, then add 80times(volume) water to the extraction-container, Add the sample when the temperature of thewater raised to 95℃. and terminate the operation 4h later in 100℃. Centrifugated the liquid, and deposited the clear liquid used 3 times (volume/volume) alcohol, then filtrated thedeposition after 12h. dry it in 50℃. At last, crush it up and airproof it.The experiment of the antivirus activities in vitro was carried out used thepolysaccharides, obtained by the technics of hot water extraction, microwave-assistedextraction and magnified experiment, and the Carrageenan which obtained from the market.Results showed that the polysaccharides obtained by the technics of hot water extraction, andmicrowave-assisted extraction had a good antivirus activity while the Carrageenan obtainedfrom the market did not. It also confirmed that the technics of microwave-assisted extractioncould protected the biological activities of the polysaccharides effectively, and the feasibilityof the magnified test. |
